While preparing for COMLEX, you need to study not only hard but smart. The first step is the most difficult one, and you will need to work diligently to overcome it.
You need to have study materials for Step 1 of the COMLEX. Everyone has a different method of studying. Some like quiet reading, others like to read aloud, and some prefer to write down things as they read. Even others learn by interacting and having the hands-on experience with the subject. Use whatever method suits you, but be sure you have the right materials for the job.
You need to use a combination of textbooks and online resources to study. Some important books to purchase are:

The National Board of Osteopathic Medical Examiners (NBOME) is in charge of the exam.
You can take a practice exam on the NBOME site at: http://www.nbome.org/practice/level1/index.htm
These are online sites where you can take exams like the COMLEX. Be sure to take notes and time yourself while taking these practice tests. Time is a major factor and plays a crucial role during the exam. You should listen to the Goljan audio lectures to get more information.
You have to take the Comprehensive Osteopathic Medical Self-Assessment Examination (COMSAE). The COMSAE is a good alternative to the USMLE. It is extremely helpful for anyone who wants to take the COMLEX. The question style, structure, and format of the COMSAE is almost identical to that of the COMLEX. You can familiarize yourself with the exam and build confidence.
Try not to learn by rote as the question may show up in a different form on the exam. Learn about topics and spend time researching questions for Step 2. Some Step 2 questions appear in Step 1, and it helps to be ahead of the
