Lennie is played by John Malkovich. Malkovich’s portrayal of Lennie has been intelligently thought out, from the physical size which emphasizes Lennie's brute strength, to the contrast in his manner of speaking, which is slow, deliberate and filled with constant stuttering. Averting eye contact, and Lennie’s repetitive furrowed brows allow the viewer to visually connect Lennie’s innocent mentality with his brute strength.

Gary Sinise, who plays George, portrays his sharp tongue in dealing with Lennie through persistent eye contact and sharp responses to Lennie’s behavior; which exemplifies his quick-temper and lack of patience.
