Dear Members of the Review Committee:

I am writing this letter on behalf of Neil Chappell, MD, in order to convey my strong support of his grant proposal entitled, “Hyperandrogenemia alters ovarian mitochondrial function and subsequent oogenesis and embryo development in a PCOS mouse model.” As his mentor and principal investigator, I am committed to the development of his career, and will assist him in any way I can to ensure his success in all professional endeavors. To date, Dr. Chappell has accomplished many significant achievements, and I believe in his capacity to continue to grow as a clinician scientist to become a prestigious example in this field as a well-respected independent researcher.
Dr. Chappell completed his medical school training at University of Texas Health Science Center at Houston, and subsequently completed a four year residency program in Obstetrics and Gynecology at the University of Alabama at Birmingham. During his training, he regularly received awards and recognition for his academic achievements. While in medical school, he illustrated robust interest in the field of reproductive endocrinology, earning the Outstanding Medical Student in Obstetrics and Gynecology and the National Endocrine Society Medical Student Achievement Award. In residency, he was

Chappell as his mentor in his research project on the role of androgens on the mitochondria and subsequent oocyte and embryo development. It is an area of science that warrants much study, with vast implications to expand our knowledge, not only of basic ovarian physiology, but also of the many phenotypic presentations of polycystic ovary syndrome. With his career plan in place, Dr. Chappell stands to become an expert in this realm, and has the resources available to him to better illuminate the finer points of this disease, and thus cultivate novel treatment paradigms that are better suited to this complex

Occasionally these cysts form in small clusters that contain immature eggs, and are often associated with polycystic ovarian syndrome. PCOS is a hormone imbalance problem believed to involve increased insulin levels that cause the ovaries to produce excess androgens, which hinders ovulation (Eisenberg, 2014). There is no known specific cause of PCOS, but research shows that some women are more likely than others to develop other diseases or conditions as a result. Women with PCOS have a greater chance of developing high blood pressure, high cholesterol, sleep apnea and diabetes (Eisenberg, 2010). In particular, African American women are just as likely to develop PCOS as white women, but have a higher likelihood to develop more severe symptoms, develop diabetes or experience weight gain or obesity (Ladson,…

Introduction The percentage of men over 40 using testosterone replacement therapy (TRT) tripled from 2001 to 2011¹. Newer, gel based forms of treatment have aided the ease of application and encouraged prescription rates. Along with this trend comes an increase in the data surrounding TRT. This influx adds to the large scale, randomized studies conducted on the use of TRT in hypogonadal men. TRT is widely accepted to provide relief from retarded spermatogenesis, low sex function, depression, and bone fiber deterioration in aging men.…

Pregnenolone serves as a principal precursor to all the other steroid hormones synthesized by the ovary, testes, or adrenals. It shows that the rate-limiting step for the production of progesterone is side-chain cleavage of cholesterol by P450scc. Pregnenolone is then converted into progesterone by 3β-hydroxysteroid dehydrogenase (3β-HSD). The deficiencies in either P450scc or 3β-HSD have ardent effects on the synthesis of all steroids. In the ovary, progesterone is produced at all stages of follicular development as an intermediate for androgen and estrogen synthesis but becomes a primary secretory product during the peri- and postovulatory (luteal) phases.…

Also, cell networks can be regulated by hormonal mechanisms because gonadectomy alters the responsiveness of cells to GHRH, including hormone release and cell motility (71, 72). In gonadotropes, GnRH treatment leads to an increase in process extension, which is enhanced by prior E2 exposure (73). Therefore, it is evident that sex differences can exist in pituitary networks and these networks can be regulated by gonadal…
