Introduction
Antibiotics are the natural substances produced by the microbes that destroy or slow down the growth of the bacteria, whereas antimicrobial agents are the synthetic substances which developed in the laboratory that mimic effects of antibiotics. Selective toxicity means the microbial agents that kill the microbes without killing the host cells. Antimicrobial is divided into two spectrum of antibiotics –
1 Narrow spectrum: - This antibiotic is active against a selected group of the bacteria and will cause less resistance (SRS. n.d)
2 Broad spectrums: - This antibiotic is effective against a wide range of infectious micro-organisms which includes both gram positive and negative bacteria. (SRS. n.d)

-Antibiotic disk of gentamycin, novobiacin and penicillin
-Nutrient agar in petri plate.
-Metric ruler, markers and plastic tweezers.
-Tea candle light, sterile swab, rubbing alcohol and bleach
-Other includes personal protective equipment like an apron, gloves, and safety goggles.
Kirby Bauer diffusion is widely used test for antibiotic effectiveness in certain kind of organisms. The steps I followed to complete this test are-
- First collect all the materials then wash the hands thoroughly and put the personal protective equipment.
- Clean the working surface to prevent from cross contamination.

- Take out the petri plate with nutrient agar in it. Divide the petri plate into three equal sections with permanent marker and label the antibiotic name (gentamicin, novobiacin and penicillin) in each section.
-Remove the lid of the active S epidermis broth, flame the lip of the tube with help of the tea light candle and careful remove the sterile swab and submerge the swab into the active S epidermis broth. -Swab the entire agar plate including the edges and let the active broth absorbed in the

Introduction: The effectiveness of Antimicrobial agents against Microorganisms Population of microorganisms or growth are killed and/or inhibited by natural or synthetic chemical antimicrobial controls (Madigan, et al 2012). Antimicrobial agents are grouped according to their efficacy against microorganisms. Bactericidal that kill and bacteriostatic prevent and/or reduce growth (Madigan et al, 2012).…

I measured 11mm which translated to being susceptible (sensitive to bacitracin). Due to this test, looking at the key I was still on the left side with it saying >10 and susceptible. Nitrate Reduction was the next test, which ended up being a very tricky one. This test had two periods, the first was identical to all the others. Obtain a tube, this one contained nitrate broth, label the tube and then aseptically inoculate the tube with the unknown bacteria.…

Furthermore, because of this issue a culture is also needed to detect resistance to antibiotics. For a culture test a endocervical or urethral swab specimen is needed. Specimen can also be taken from the rectum and pharyngeal. The specimen is streaked onto a Thayer-Martin or Martin Lewis selective medium. It can also be streaked onto a chocolate agar nonselective medium and incubated at thirty-five Celsius to thirty-six Celsius and with a five percent of carbon dioxide.…

Determining the cell shapes will help with the process of elimination organisms. Under microscopic examination for my unknown 16 broth, it appears to be a coccus shape and a gram positive. Gram staining can determine cell morphology, size, and arrangement. In a positive gram stain, a primary stain called crystal violet is used to stain the cell. For every new stain, mordant, decolorizing agent, and counterstain, it must be rinsed with distilled water.…

The lab instructor issued out a test tube labeled with letter ‘I’, which consisted of two unknown bacteria, Gram-positive or Gram-negative that were streaked from a pure culture. Sterile techniques were followed while performing precise instructions as stated in the referenced Laboratory Manual. Example 1: The first procedure performed was done by isolating a pure culture from the mixture onto a solid Trypticase soy agar (TSA) media. Sterile technique was done by flaming the loop until it turned red to ensure that there were no current bacteria on the loop avoiding contamination followed by rapidly flaming the neck of the test tube to prevent the entry and contamination of unwanted microbes.…

After transfer is completed, student can begin working on their first step of biochemical testing. The first procedure in the biochemical tests of enteric bacteria was to use a bunsen burner to sterilize the inoculation loop then streak unknown bacteria on to the MAC agar plate using streak plate technique referred from the lab manual in exercise 7. The MAC agar plate was then stored in the incubator at 37°C for about 48 hours until the next lab time.…
