The Integumentary System is the largest body system of humans. Being the largest there can be a larger number of possible issues, than other body systems. Skin cancer is a growing issue in this country. Cutaneous Squamous Cell Carcinoma is one of the most common forms of skin cancer. Our very own sun can cause this cancer, also causing it to advance from the Ultraviolet Rays. In studying Cutaneous Squamous Cell Carcinoma (SCC) through DNA Methylation profiling, two subclasses of SCC have been found to have two distinct cell origins. Actinic Keratosis (AK) are UV-induced precancerous corruption of cells in the skin. The investigation of AK and the way it turns into SCC isn’t completely comprehended. Research shows that AK methylation patterns display standard features of cancer methylomes, being very similar to SCC. ” SCC is a non-melanoma skin cancer type that originates from epidermal keratinocytes and represents about 20% of all skin cancers, with up to 700,000 new cases annually diagnosed in the USA.” (page 2; paragraph 1; line 1-4). There are some obvious transcriptional networks transferring AK from the healthy side to the cancerous side. “DNA methylation is a covalent epigenetic modification of cytosine within CpG dinucleotides.” (page 2; paragraph 2 line 1&2) These modifications use enzymes known as DNA methyltransferases to normalize cell identity through gene expression. One recent study showed UV- irradiation from deep-rooted sun damage produces large amounts of hypomethylated blockage in healthy epidermis preserved in SCC and the suppressing of miR-204 may play an important role in the advancement of AK to SCC.
