Squamous cell carcinomas of the lungs, head and neck, esophagus, and cervix account for over 400,000 deaths per year. Finding a way to effectively treat this type of cancer is very important for the well being of people. In this primary research article, the authors attempt to find a mechanism by which squamous cell carcinoma proliferation and invasion are inhibited. They use the cell adhesion molecule 1 (CADM1), which is a tumor suppressor, to show that it is possible to prevent squamous cell carcinoma progression. One result that could come from this study is that we might be able to screen the patient’s tumors for loss of CADM1 expression to see if they will benefit from JAK/STAT targeted chemotherapies. To find out if CADM1 expression did in fact inhibit squamous cell carcinoma progression, they studied its effects on the growth and migration of the CADM1-null squamous cell carcinoma cell line A431. Some of the cells were treated with doxycycline. The CADM1 transduced cells that were treated with doxycycline restored CADM1 gene and protein expression to normal levels. The CADM1 transduced cells that weren’t treated with doxycycline could not be distinguished from the squamous cell carcinoma controls. The researchers also used mice in their study. They injected the mice with either control or CADM1 expressing A431 cells. Reduced squamous cell carcinoma growth was seen in CADM1 expressing tumors compared to controls, and also significantly fewer squamous cell carcinoma metastases were seen in A431-CADM1 tumor bearing mice compared to those seen in untransduced squamous cell carcinoma controls.
