Escherichia's Growth

Decent Essays
This is the result of Escherichia coli after a week of incubation and being placed at room temperature for ten minutes prior to being incubated. This was expected to show the most amount of growth because pathogens will grow faster at 37°C, the temperature of the human body. This is evident by the constant lines of colonies of the plate. Incubation temperature is 37C, which is optimum temperature for its growth. Mesophiles, like Escherichia coli, are bacteria that thrive at this temperature because it is able to grow and divide very rapidly. The plate is not countable because there are over 300

If it is examined within 24 hours of plating it will demonstrate alpha-hemolysis, but if examined past 24 hours it will demonstrate gamma-hemolysis (2). Unknown 15 was examined after a one week incubation period, thus, it demonstrated gamma-hemolysis. Mannitol Salt Agar (MSA) was used to distinguish whether Unknown 15 belonged to the genus Enterococcus or Lactococcus. When streak plated onto the MSA medium, Unknown 15 grew on the plate and produced a yellow color change. The growth and color change (indicative of mannitol fermentation) resembled the the positive control Staphylococcus aureus.…

I obtained my TSA plate from the incubator and determined that there were two separate species present due to the size of the colonies. One species had very small colonies and the other species had larger colonies. I inoculated each different species with the aseptic loop technique and made two separate gram stains to determine the gram reaction and shape of the two separate species. I found that the bacteria that formed the smaller colonies are gram negative bacilli, and the bacteria that formed the larger colonies are…

The first step was to streak the bacterium with a sterilized look onto a Mueller Hinten plate. The MH plate was incubated at 37 degrees Celsius for 48 hours. Upon return and observation, the MH plate did not isolate the colony enough to be able to test the bacterium. The professor then handed an alternate 10 that was a pure culture to streak. It was with a sterilized loop that some of the pure culture was taken out and gram stained as directed by the laboratory manual (1).…

The purpose of performing physiological tests is to figure out certain characteristics of microorganisms or to reach a conclusion on what an unknown microorganism is. It can be also used to determine what is needed for bacteria to be cultured in or on certain media. Culture media can either be liquid or solid, and they can be used to identify certain characteristics of different bacterial species. This is indicated by the growth or lack of growth in or on certain media. Solid media can also be used to isolate organisms and provide a means to observe their morphology (Wiley 55).…

[2009], where strain to strain differences in the growth and survival of various V. parahaemolyticus and V. vurnificus were observed at refrigeration temperatures. A well-known strategy for modeling is to choose the fastest growing strain in the environmental conditions investigated, because the fastest growing strain will dominate the growth in food products (Rash et al., 2004). McMeekin et al. (1993) also recommended independent modeling of several different strains before choosing the strain that grows fastest under the environment conditions of most…

al’s (2015) study, with a clearly indicated optimal temperature range around 70° C or higher. However, the process of denaturation likely would not have been indicated by results due to the use of bacteria as opposed to fungal high-fiber/protein biomasses that denatured around 95° C in George et al.’s (2015) study. Serrano and Peralta’s (2015) study provided evidence that the conditions of an organism’s natural environment influence its enzymatic activity. Thus, it is likely that bacteria would typically have higher rates of enzyme activity present at higher temperatures as compared with other living organisms. If precautions were taken and the experiment was replicated, it is likely that the results would further support George et al. (2015) and Serrano and Peralta’s (2015) findings.…

This section will be a narrative summary of the characteristic of my unknown bacterium that’ll touch base on the morphology, cell well structure, temperature tolerance, antibiotic resistance, physiology and other interesting experiment results. First is going through the morphology of my unknown bacterium; will be talking about the shape, cell wall structure, and other characteristic using the results from tests such as gram staining, simple staining, and spore staining. According to my lab notebook, the result from gram staining of my known is seen to be a rounded shape, cocci shape all bunched and clustered together like a bunch of grapes at 100x using the microscope (Kaddo, 2017, pg. 22). The color after the safranin wash from gram staining…

Escherichia Coli is Gram-negative rod; motile; with or without capsule; non-fastidious, facultative anaerobe; bile tolerant; capable of growth at 44°C. Normal habitat is gut of humans and animals; may colonize lower end of urethra and vagina [1]. Most strains of Escherichia coli are common microflora in the human and animal gut flora. However, a few strains are potential foodborne (and occasionally waterborne) pathogens and produce potent enterotoxins [1,2].…

In today’s society were one can get food from an abundant amount of sources, it is very important for the person to know where the food they are consuming came from. Many Americans are trying to eat a healthier diet consisting of fresh and raw products. They are trying to be more aware of what they are putting in their bodies and are shopping more at farmer’s markers, natural food stores, and purchasing more organic foods or even growing their own produce. In addition to that, the people are trying to eat foods that contain less additives in them along with foods that have minimal processing to them. This is great that more people are concerned about their health and want to do what is best to insure they are healthy, however, some of the…

Although there was food present for the bacteria, the ampicillin contradicted with growth and reproduction. The fact that there was no DNA plasmid had a role in the bacteria not glowing. Since there was no plasmid on the plate, the gene for green fluorescent protein was not even present, so there was no way for the bacteria to glow. The hypothesis about the -pGLO plate was correct because the bacteria ended up growing but not glowing under UV light. The bacteria had such a prominent growth rate…

Follow the same steps until all three quadrants are completed, but avoid entering quadrant one only take from quadrant two. The purpose of the streak plate method is to produce isolated colonies of an organism. The TSA plate was incubated for approximately 48 hours at 37 degrees simply to grow bacteria. After returning back to the laboratory and noticing growth on the TSA plate the same streak plate method was applied to the MacConkey agar and the Mannitol salt agar.…

When we think about ice, snow or rain falling from the sky, we imagine some perfect beautiful crystals made of water molecules. Scientist had discovered that those crystals originate because some dust particle, helped with the wind, goes to clouds and then water surround it. Because high in the atmosphere the temperatures are so low, those water molecules freeze giving a structure called ice crystals. Now we know that precipitation from the sky are much more that simple dust and water.…

Listeria monocytogenes is a gram-positive rod-shaped bacterium and facultative anaerobic that can grow under either anaerobic or aerobic conditions but do not form endospores. Among the six species of Listeria, only Listeria monocytogenes is the major human pathogen that causes disease in humans. It is a causative agent to cause listeriosis, a serious infection caused by ingesting the food contaminated with Listeria. The optimum temperature for growth is 30 to 37 degree Celsius, within this range, they multiply the best. Also, they can multiply better than all other bacteria at the refrigerator temperatures.…
