When a blank containing boiled (denatured) enzyme instead of an active enzyme could be used but it would not be the better blank to use in this experiment. The better blank solution would have been the one with active enzyme because it would cause the spectrophotometer to measure the absorbance of every component in the blank except the substrate, creating a 100% transmittance. This allow the spectrophotometer to read everything that was not in the blank when the actual sample is inserted in because it would “blank” out all the components in the “blank” solution and only read the “uncommon” component. With the active enzyme, the spectrophotometer would “blank” out the enzyme and only read the substrate absorbance because according to the lab …show more content…
To increase the concentration of the substrate, leave the concentration of enzyme constant and add more volume of substrate in different tubes and then test of the absorbance values. Then using the standard curve, convert the absorbance values to concentrations and then plot them to look for a trend of increase. For example, set different increasing amount of substrates like Tube 1 would have 0.5mL, Tube 2 would 0.6mL … etc and see which volume of substrate would cause an increase in the production of …show more content…
A major area where enzymes are present a lot in is the food industry. A lot of the food we eat contain enzymes. Lactase is an enzyme that a lot of people know about. It cuts lactose into its smaller sugars: galactose and glucose. When the small intestine of a human does not produce a sufficient amount of this enzyme, this condition is known as the lactose intolerant, which is when a human has discomfort in the form of cramps, gas and diarrhea when they eat/drink milk products. Lactase is also used to make lactose-free products for people with this condition. Many other enzymes are present in the foods we eat and they all have different functions. These enzymes include hemicellulose, lipase, and amylase which are used in the baking industry along with proteases and carbohydrases which are used in the meat processing industry. Enzymes are also used in the medicine industry. Enzymes can be used to treat disorders such as “low blood pressure, myocardial infarction, tachycardia, head or spinal injuries, there are chances of formation of blood clots due to stagnation of blood inside the blood vessels,” enzymes are used to break down the clots so blood can flow better. Digestive enzymes like papain are given for patients to take orally after eating to make for easier digestion. In humans, there is an enzyme called Acrosome present in sperm to help them penetrate the uterine tissue so

INTRODUCTION: The objective of this lab is to measure the activity of an enzyme and the effects of environment conditions on enzyme activity. Enzymes are catalysts; agents that speed up chemical reactions by lowering the activation energy required. This means that a catalyst helps reactions occur at a greater speed and lower temperature.…

We first set up a blank to zero out the spectrophotometer. In tubes 1-4 we added 200 ul of DPIP and 200 ul of the bean extract enzyme, but in tube two, the bean extract was boiled. We then added the appropriated amount of phosphate buffer to the tubes. 200 ul of the inhibitor, malonate, was added to tube three and four. 66 ul of the substrate, succinate, was added to test tube one, two, and three but 200 ul of succinate was added to tube four.…

Liver Lab By: Jonah Stillman Background Information: Catalase enzymes are inside of nearly every living organism that is exposed to oxygen. Some examples of catalase enzymes are bacteria, plants, and animals. The main job of a catalase enzyme is to catalyze the decomposition of hydrogen peroxide to water and oxygen. When an enzyme is exposed to hydrogen peroxide or certain other chemicals it is prone to react. Catalase enzyme reactions also depend on the temperature they are exposed to.…

Catalase Enzyme Lab Write-Up Our goal with this lab is to discover how enzymes affect the production and destruction of harmful chemicals our body makes into harmless chemicals. Our body contains hundreds of enzymes in our cells and this lab will show us how they interact with parts and materials of our bodies. The enzyme we will be focusing on is called catalase. It’s job for the body is to break down hydrogen peroxide a harmful substance into two harmless substances oxygen and water.…

They catalyze most of the reactions compromised in metabolism and they do not just make the majority of the reactions possible, but also serve as mechanism of stabilization and control for these reactions. It is fair enough to admit that without the presence of enzymes life would not be possible. Enzymes kinetics focuses on how the enzymes behave in response to different concentrations of both substrates and products. During this lab the kinetics of the enzyme alkaline phosphatase will be explored. Alkaline phosphatase is considered as an ubiquitous enzyme which can be obtained by isolation from kidney, bone, plasma, intestine, liver, among others.…

An enzyme is a protein catalyst, whose basic function is to increase the rate of the reaction, meaning lowering the activation energy. Because the body creates enzymes to be as specific as possible, in order to prevent inefficiency and careless mistakes, most enzymes come in direct contact with only one substrate to produce products. Enzymes will work on creating substances until the reaction is complete or the particular enzyme is inactivated, which either means being destroyed & recycled or paused, for the essential enzymes needed on a regular basis. An example of a necessary enzyme is catalase, better known as peroxidase, which catalyses or decomposes the reaction H2O2 (hydrogen peroxide).…

Enzymes are true catylst that exist in the living matters, helping to speed up the reactions and to keep up the metabolisms within the cell. Enzymes will work properly when working in favorable condition and with substrates that will bind easliy to form products. Enzymes bind to specific sustrates for form specific product(s) in different parts of the cell. These happens in the active site where chemical reaction begain as substrates are binded to the enzymes. Enzymes have different shapes, bond attractions or size that will fit to particular substrates and may also cause configuration changes within the enzymes that produce products.…

Lactose is a sugar found in milk and milk products. Lactase Is an enzyme found in the small intestine that breaks down lactose into simple sugars such as glucose. The substrate has to fight to bind with the enzyme lactase. The inhibitor tries to attach to the lactase and slow down the chemical reaction while the substrate does the opposite. The substrate increases the rate of the chemical reaction, this chemical reaction breaks down the lactose into sugars.…

Exploring Enzymes Dmitri N. Piquero Biological Discovery I Laboratory Section I Abstract Amylase is an enzyme that speeds up the conversion rate from starch to glucose. Four test tubes were filled with a starch solution and a different concentration of amylase solution ranging from 0 μL to 1000 μL. The test tubes were then left to soak in a 37°C water bath before having 2 mL of 1 M hydrochloric acid added to a test tube of pure amylase solution. Lugol’s iodine was used to test for the existence of starch in the samples, while Benedict’s reagent detected any broken-down glucose.…

Enzymes are found in all living organisms. They are present to speed up or accelerate a chemical reaction; referred to as a catalyst in a chemical reaction. They are essential to obtain rates to sustain life as without enzymes rates would be too slow to provide for life. Molecules at the start of the process are called substrates, the enzyme will then convert these into changed molecules which are called products, after this process the enzymes maintains the same shape. (Berg et al. 2002).…

Introduction: Lactose intolerance is one of the most common conditions, which affects millions of people. People with this condition do not produce the lactase enzyme which metabolizes the sugar on their own therefore they are unable to break down the Lactose which is sugar that is found in milk. Lactase is an enzyme found specifically in milk sugar “Lactose”. It breaks down the lactose into monosaccharaides, galactose and glucose. The Lactase enzyme can be used to aid people with this condition.…

As enzyme concentration increased, the color strength continued to increase. In Biology in the Laboratory, this is confirmed by the section concerning the effect enzyme has on activity (Helms et al,. 1998). In Figure 4, the substrate and color relation had a direct relation to an extent. As substrate concentration increased, the color strength continued to increase until the solution reached maximum velocity.…
