EXPERIMENT: The effect of an acidic fluid on the activity of an enzyme
INTRODUCTION: There are several factors that may influence the activity of an enzyme to include temperature and the pH levels. In order to better understand how the pH level affects the actions of an enzyme, the experiment will introduce different foods with acids and bases through mixture of direct contact.
OBJECTIVE: The objective of this experiment is to recognize how the introduction of an acid affects the activities of an enzyme.
MATERIALS REQUIRED: Meat, acidic foods, potatoes, alkaline foods, a scaled beaker, yeast, boiling tube, hydrogen peroxide, 100 milliliter graduated cylinder, and a dropper
HYPTHOSIS: If the pH level can affect the activity of an enzyme, then

Utilizing an acidic food and an alkaline food will yield how the pH and the activity of the enzyme is effected on the acidic foods. With the catalase that is present in the yeast, the oxygen was released in bubble-form. By measuring the bubbles, the action of the enzymes can be easily seen. The experiment followed these steps:
1. One centimeter cubed meat added to the 100 milliliter graduated cylinder
2. Two centimeters-cubed of the source of the catalase enzyme, yeast, was added into the 100 milliliter graduated cylinder
3. Two milliliters of the hydrogen peroxide were put in a boiling tube
4. The graduated cylinder and the boiling tube were both placed into a water bath that had a temperature of 82 degrees Fahrenheit
5. Once they were at the same temperature, I poured the hydrogen peroxide into the graduated cylinder. At the same time, I noted the overall initial volume of the of fluid in the graduated cylinder
6. After waiting two minutes, I made note of the volume of the fluid in the graduated cylinder
7. I subtracted the initial volume from the final volume in order to determine the change in

INTRODUCTION: The objective of this lab is to measure the activity of an enzyme and the effects of environment conditions on enzyme activity. Enzymes are catalysts; agents that speed up chemical reactions by lowering the activation energy required. This means that a catalyst helps reactions occur at a greater speed and lower temperature.…

Solutions with a pH of 4(acidic), 7(neutral), and 10(basic) will be tested with the tyrosinase and catechol. Each solution will be tested twice for validity. The solution with the same pH as water will produce a better environment for the enzyme to react. This reaction was previously proven in experiment…

From 10oC to 20oC, the average rate of reaction of the class results is higher than the raw results rate of reaction by a maximum of 0.75mL/sec. However, from 30oC to 50oC, the raw results presented a slightly higher lot of rate of reaction than the average rate of reaction from the class results. The highest rate of reaction difference was for 50oC, with a difference of 0.3mL/sec between the raw and average results. This information supports the hypothesis, yet the result for 10oC and 20oC do not support the hypothesis, as due to the information known about enzymes, the lower the temperature, the slower the rate of reaction should be, therefore the average rate of reaction does not support the…

The hypothesis for the: Chicken Liver in Room Temperature: The hypothesis was that there will be a reaction. The chicken liver enzyme catalase only works when the liver itself is at a stable internal temperature. Chicken Liver in a Boiled Temperature: The hypothesis was that there will not be a reaction. Due to the chicken liver being in a boiling environment, the enzyme in the liver will have already been denatured. Chicken Liver in an Acid environment: The hypothesis was that there will be a reaction.…

The experiment was carried out at different temperature (27° C, 32° C, 37° C) and pH (4, 6 and 8). Different sources of carbon such as maltose, sucrose and glucose at 1.5 % (w/v) were used and nitrogen sources such as beef extract, peptone and yeast extract at 0.3 % (w/v) were used to determine their effect on the production of protease…

Temperature and pH will affect enzyme function by increasing the reaction rate because as temperature increase so is the rate of reaction and extremely high or low pH will result in the denatured by decreasing the reaction rate. The function of this experiment was to investigate the influence of the concentration of enzyme on the rate of reaction. The rate of reaction in this experiment was subjected by the amount of Guaiacol, H2O2, Extracts, and Buffer (pH5). The reaction rates with different enzyme concentration at 0.5x (ml) was slow but gradually increase making the absorbance below 0.1NM. At 1.0x (ml) the rate was a little slow but then it increased rapidly.…

In the test tube #1, there was a few bubbles compared to the tube #2. The difference in the reactions was in test tube #1, the catalase was denatured by the high temperature, but in test tube #2, there was no denatured occurred, so the catalase could convert hydrogen peroxide into water and oxygen. The optimal condition for catalase in this experiment is the temperature should be not too high, so the catalase can’t be denatured. In the experiment 4.4, we could see the effect of pH on enzyme activity by adding 2ml hydrogen peroxide and a small piece of liver to each test tube and adding 20 drops of acidic solution into tube #1, adding 20 drops of dH2O (pH 7) into tube #2, and adding 20 drops of basic solution into tube #3. We observed that the test tube #2 had the greatest bubbles, and the second one was the test tube #1, the third one was the test tube #3.…

However, when the substrate concentration was increased, the intensity of color stopped increasing due to the enzymes being unable to to process at a faster rate. All of the alternate hypothesizes were supported; in the experiments, section one (with Figure 1) had an optimal temperature at approximately 24°C; section two (with Figure 2) had an optimal pH at approximately pH8; in section 3 (with Figure 3) the color continued to increase with the addition of more enzymes; and in section 4 (with Figure 4) the color continued to increase until it reached a maximum velocity with the addition of more substrates. One desired retrial for the experiment would be to test for more points after 20 drops of potato juice in Figure 3. Understanding the effects of how these properties alter the ability of the catecholase enzyme could allow for better insight into decreasing time needed for biochemical reactions, proper food storage, and the condition of…

First, students add a drop of water into the test tube with no hydrogen peroxide in order to display the water’s ability to sustain the reactants and products within the substance. It is also important for students to correctly order the chemicals when inserting them into the test tubes. Hydrogen peroxide is added first, which is decomposed and processed into oxygen gas by water and active yeast. H2O is then added in order to control the level of hydrogen peroxide in the substance. Finally, chicken yeast is added in order to speed up the decomposition reaction of the H2O2.…

Introduction: Enzymes have a huge effect on the human body and other organisms. Enzymes are catalysts that are used to accelerate the reaction process by lowering the activation energy. For this experiment the catechol oxidase was extracted from potatoes. The enzyme is called catechol oxidase because the oxygen is reacting to catechol. Catechol oxidase causes the browning of the fruits and vegetables because the compound is toxic to the bacteria.…

Again, there was not a difference in tubes four and five in foam height. My conclusion is that when you have hydrogen periodic and potato juice the lower the temperature, the higher the catalase reaction will be. Because test tube two was not far behind test tube one when it came to foam height, this study raises the question of whether there might be an optimum temperature in enzyme activity. On the basis of the results of this study, the hypothesis presented can be taken in to account, and be accepted. Additional experimentation with different temperatures is suggested.…
