A Possible Mechanism for Blindness Associated with Diabetes: Na+-Dependent Glucose Uptake by Retinal Cells The results displayed are indicating that there might be a possibility of a sodium-coupled glucose transporter (SGLT). By interpreting the results, one is able to see that the glucose uptake is higher when sodium ions are present than without them in the pericytes. Glucose uptake on endothelial cells does not change whether sodium is present or not. The plotted results are displaying two distinct trends. As sodium concentrations increase, glucose uptake by retinal pericytes would increase as well. A hyperbolic curve is formed from these results and appears as if the glucose uptake begins to settle as the concentration of sodium

The Km tells us the substrate concentration when vmax is halved. With this information we are able to see how efficiently an enzyme efficiently converts a substrate into product.
Investigators have convincingly demonstrated that pericytes have SGLT since the glucose uptake rate is dependent on the amount of sodium present. Endothelial cells on the other hand do not have SGLT since their glucose uptake is unaffected by sodium concentration. a. When inhibitors such as galactose and 2-deoxyglucose are used, the glucose uptake slightly decreases in both the presence and absence of sodium.
b. When phlorizin is added the transport, it regulates the glucose uptake levels with a noticeable decrease of glucose uptake when sodium is present. When sodium is absent, the phlorizin does not seem to create much of a difference when compared to the control. As represented in the data, both types of collagen (Type IV and VI) would increase when glucose concentration levels and the cellular density increase. With the presence of phlorizin, glucose consumption is halved in both the low and high concentrations. A likely cause for the great amount of reduction in the higher concentration might be partially due to how reactions work. When higher concentrations are present then the reaction rate is increased.
b. Collagen synthesis is reduced in the presence of phlorizin in both cases of high and low concentrations, but there is a notable difference when a higher concentration of glucose is present. The higher concentration of glucose may be responsible for the great amount that was decrased. Retinopathy is more likely to occur in a diabetic patient since glucose problems will be present. In the case of a patient with Type 1 diabetes, a patient will have trouble producing insulin therefore glucose levels would be high when unregulated. Although a patient with Type 2 will be able to produce insulin, they will have trouble processing glucose and will have high sugar levels.
