Cyclin D1 Lab Report

Summary
The overall goal of this experiment was to determine if there is a connection between cyclin D1 amplification and breast cancer therapy resistance. This experiment was motivated by the statistic of forty percent of endocrine therapy breast cancer patients being resistant to the treatment. In some research, it has been thought that the presence of cyclin D1 causes resistance to tamoxifen therapy, while others do not think cyclin D1 is relevant to the efficacy of the treatment. This experiment will further the understanding of this protein and the relativity of it by performing tissue arrays of 147 breast cancer patients. It is important to find out what is causing this resistance so changes can be made and hopefully, a successful breast

It was performed multiple times on each tumor at different parts of the tumors using FISH technology. This technology used a specific stain to fluoresce the cyclin D1 gene in one color (pink) and the centromeres of chromosomes as another color (green). Cyclin D1 amplification was present when there was a large amount of pink stain when compared to the amount of green stain, and was not present when the two quantities of the colors were relatively equal. As a result, there was only about 21% of the cancers that showed cyclin D1 amplification. It is thought that cyclin D1 amplification is an early event in the development of breast cancer due to the results concluding that the degree of amplification is highly …show more content…
It states that the purpose of this experiment was to quantify the cyclin D1 amplification and that it may have an impact on therapy resistance but does not seem to have a specific side that it is trying to prove or disprove and does not list further questions that are present that make this an important topic. This is a very important topic so more detail could have gone into trying to specify why their hypothesis is important to address. They spent a little too much time talking about previous research, rather than establishing their purpose and hypothesis for further evaluating the specific topic on cyclin

Interactive Question 7.2 Cite some experimental evidence that indicates that membrane proteins drift. A good form of experimental evidence is Fluorescence Recovery After Photobleaching, aka FRAP. In the FRAP process, membrane proteins are labelled with a green fluorescent protein, and part of the plasma membrane is “bleached” with a laser, causing them to lose their fluorescence. The part that was bleached will eventually become fluorescent again, as molecules drift in by diffusion.…

In the four-week long experiment conducted in the Bio 13 Lab, we were able to conduct a genetic analysis of the yeast S. cerevisiae, particularly investigating the inheritance and linkage of three genes: HIS4, LEU2, and LYS2. In week one, a suspension of spores was received and aliquots of the suspension were plates onto two YPD plates. After a week of yeast growth, haploid spore clones were carefully picked from the plates by sterile toothpicks and used to make patches on a new YPD plate, the master plate. In week three, the master plate was replicated onto three dropout plates: -His, -Leu, and -Lys. In the final week, the growth of the patches on the dropout plates were analyzed and class data was collected in order to conduct calculations for…

Assignment #2: The Replication of DNA in Escherichia Coli Due Date: Feb 8, 2016 at 9:00 AM CST 1a. A procedure in which a small amount of DNA material is immersed in concentrated solution, cesium chloride (CsCl), and soon after placed in centrifuge until the point where equilibrium is achieved is called density-gradient centrifugation. Contrasting techniques involving diffusion and sedimentation form a persisting CsCl concentration gradient. Furthermore, the concentration gradient and pressure of centrifugal force results in increased density.…

Magnesium start to turn into a white liquid form. After the bubble start to disappear, more Hydrochloric acid was added, and the reactions repeats. Until none of the shiny silver Magnesium ribbon can be seen, and there is no more bubble, the reaction is over.…

The control, however, is very important in the determination of possible metastases because it contains wild type (normal) DNA that can be compared to Valerie’s DNA samples. In Valerie’s peripheral blood and normal breast tissue samples, a copy of the wild type DNA is present in addition to a copy of the cancer DNA, thus she is heterozygous in this area. In Valerie’s tumor tissue sample, however, there is no copy of the normal gene present, thus she is homozygous for the cancer DNA in this area. Because Valerie has one copy of the wild type…

This is two week project is to evaluate the stereochemical outcome and facial selectivity of an oxidation-reduction interconversion between 4-tert- butylcyclohexanol and 4-tert-butylcyclohexanone while investigating the stereoselectivity of different reducing agents. During the first week, 4-tert-butylcyclohexanol, a secondary alcohol (mixture of cis and trans stereoisomers) is to be oxidized into a ketone, 4-tert-butylcyclohexanone using sodium hypochlorite (bleach). Thin layer chromatography (TLC) is used to monitor the progress of the the reaction. The second week of the reaction, 4-tert-butylcyclohexanone is reduced using sodium borohydride or the Meerwein-Pondorff-Verley (MPV) conditions. NMR spectroscopy is used to evaluate the stereochemical…

This week’s lab focused on a green chemistry reaction of cyclohexanol and acetic acid, resulting in the formation of cyclohexanone. The identity of the product was confirmed using an IR spectrum to determine the major functional groups of the compound. The two main functional groups that appear in the spectrum are the C-H stretch around 3000 cm-1 and the C = O stretch around the 1600-1700 cm-1.…

Introduction Chlorinated solvents are chemical compounds, primarily used today in the dry-cleaning industry. Structurally, they are organic compounds, typically containing one to three carbon atoms, arranged in a hydrocarbon chain. Single or multiple chlorine atoms are attached to the hydrocarbon chain by covalent bonds. As shown below in FIGURE 1, chlorinated solvents can be in three forms, dependent on the chemical structure: chlorinated methanes, chlorinated ethanes, and chlorinated ethenes. As the chemical structure of a chlorinated solvent becomes more complex, that compound is more persistent in the environment and thus, becomes more difficult to degrade.…

The synthesis of cyclohexanone is an example of an oxidation reaction. Oxidation reactions have an increase in oxygen or the C-O bond, and a decrease in hydrogen or the C-H bond. Secondary alcohols normally undergo oxidation reactions to produce ketones. In this experiment, the cyclohexanol will be oxidized to produce cyclohexanone. The exact mechanism of this has not yet been fully established but scientists of today created possible mechanisms that are acceptable.…

The dehydration of Cyclohexanol requires going through an acid-catalyzed elimination of water for the synthesis of cyclohexane from cyclohexanol. To get cyclohexane from cyclohexanol you must remove the cycloalkane with fractional distillation, drying with anhydrous sodium sulfate, and analyzed by GC data and the bromine test. The reaction we are trying to accomplish going from cyclohexanol to cyclohexene is a secondary alcohol going through an E1 reaction. This is the unimolecular elimination reaction in which Zaitsev’s rule applies where the most substituted alkene is formed.…

Breast cancer is one of the leading cancers that affect a myriad of people in today’s society. “About 1 in 8 U.S. women (about 12%) will develop breast cancer over the course of her lifetime.” (Breast Cancer, 2016). Some people diagnosed with this type of cancer could have the opportunity to detect it early on due to screenings or self-evaluations. However despite early detection, it does not guarantee that the cancer can be treated in its entirety.…
