Cudratricusxanthone A (CTXA), one of the xanthone, was isolated from Cudrania tricuspidata Bureau, and its anti-inflammatory, hepatoprotective, anti-proliferative activites has been studied in vitro; however its effects on osteoclast differentiation have not been demonstrated. In this study, CTXA was found to suppress RANKL-induced osteoclastogenesis, actin ring formation, and bone resorption in RAW 264.7 cell lines and mouse bone marrow monocytes (BMMs). CTXA significantly inhibited the JNK/mitogen-activated protein kinase (MAPK) signaling pathway without affecting ERK and p38 signaling in RANKL-stimulated RAW 264.7 cells and BMMs. Moreover, it inhibited RANKL-induced expression of c-Fos and NFATc1. The fruits of c. tricuspidata expanded in Korea, because various biological activities including anti-inflammatory, immunomodulatory, and antioxidant (Han et al. 2009. 164; Lee et al. 2012. 594; Cho et al. 2003. 544). Moreover, the root bark and cortex of this plant have been used for the medical care of neuritis, hepatitis, gonorrhea, jaundice, tuberculosis and rheumatism (Jung, Shin 1989. 544).
