Abstract: The enzyme called catalysis breaks down hydrogen peroxide into water and oxygen. In this lab we conducted a series of experiment to find the effect of concentration of substrate and the temperature of catalase enzyme.
Introduction: There are a lot of factors that can affect the rate of enzyme reaction such as the concentration of the substrate, temperature of the enzyme, and shape of the enzyme. In this experiment we only measure how the concentration of the substrate and temperature of the enzyme affects the enzyme reaction. The enzyme that we use in this lab is the catalyst and the substrate that we use is hydrogen peroxide. The catalyst is a substance that speeds up the reaction rate. In this lab we used the enzyme called catalase.

The concentration that will be tested are 0.8%, 0.4%, 0.2%, 0.1%, 0.0% H2O2. First add 10ml of first H2O2 concentration to the flask and then add 10ml of catalase to the flask. After adding the H2O2 to the flask always place a rubber stopper to the flask. Swirl the flask at a constant rate and record your data. Repeat the previous 2 steps for the other substrate concentration. Now for the Experiment 2 Effect of temperature on enzyme kinetics. Repeat the steps from experiment 1 but only use 0.8% H2O2, but use boiled (100°C), warm (40°C), room temperature(25°C), and ice cold(0°C) catalase.
Result: We discover that as the concentration of H2O2 decrease the reaction rate decrease as well. As is shown in graph 1 the reaction rate of 0.8% concentration the average rate was 71.659 ml per minute as compared to the reaction rate of 0% concentration the average rate for that was 0 ml per minute or no reaction at all. We also discover that if the catalase is boiled or cold the reaction rate is slower than the room temperature. As shown in table 2 the room temperature has the fastest reaction rate of 55.692 ml per minute. The table 3 shows the P- value of the temperature

The result proves that and our hypothesis to experiment one was correct. The reaction rate for higher concentration was higher because in higher concentration there are more molecules reacting. For 0.8 concentration the standard deviation was higher two groups had a higher reaction rate than the average. We were group three and we had a lower reaction rate than the average of 0.8%, 0.4%, 0.2% concentration. We had lower because we might have swirled the flask at a different pace or for some we could have started the timer
