Cannulae implantation: For stereotaxic implantation of Cannulae, mice were anesthetized with 1.0-1.5% isoflurane and secured in a Kopf stereotaxic apparatus. Bilateral stainless steel guide Cannulae (26G, Plastics One) were targeted for placement 1.0 mm above the center of the Anterior Cingulate Cortex (ACC) (AP: +1.5 mm relative to bregma; ML: ±0.4 mm; DV: −0.5 mm from the surface of the skull) or 1.0 mm above the center of the prelimbic subdivision (PL) (AP: +1.7 mm relative to bregma; ML: ±0.4 mm; DV: −1.5 mm from the surface of the skull) (Paxinos and Franklin, 2008). Clearance through the guide Cannulae was maintained with 33G obdurators (Plastics One) cut to not project beyond the tip of the guide and protected by a dust cap (Plastics One). After surgery animals were housed individually and given 5 days of recovery during which they were monitored daily to check their health condition. For each section, a 10X tile scan image was obtained and followed by image captures with 63X objective of uniformly distributed 16 positions in PL and 8 positions in the ACC of mPFC. Only projection images are shown. The fluorescent intensity was analyzed using Image J software. Three selected neurons from each 63X image, 2 positions from each region (ACC and PL) and 4 sections from each mouse brain (n=4 animals each for control and fear conditioned) were used for the analysis.
