CRISPR stands for Clustered Regularly Interspaced Short Palindromic Repeat. CRISPRs are segments of DNA in prokaryotes that contain short repetitions of base sequences and the loci range from 24 to 48 base pairs. CRISPR-associated (cas) genes are attached to the repeat-spacer array (virus DNA fragments that tried to attack the cell).CRISPR/Cas system is an immune system that naturally functions as a defence mechanism in prokaryotes to screen for and cleave specific DNA sequences. It benefits prokaryotes because it offers resistance to invasive genetic elements such as phages.

To examine the differentially expressed candidates in pancreatic ASC, I will isolate total RNA from UPF1 mutant cell lines and perform RNA sequencing. To establish whether the upregulated genes are direct targets of NMD, I will compare this data with RNA isolated from cells where UPF1 is downregulated using short hairpin RNAs. In addition, the two major tools used to study host adaptation in IAVs, serial passage in animal models and targeted mutation, have previously been used largely independently. This research is innovative because it utilizes both of these approaches, and examines an aspect of IAV biology that has been poorly studied. We have previously identified a number of adaptive polymerase mutations, and this proposal expands this work to demonstrate a link between adaptive mutations and changes in virulence.
