Purpose:Bortezomib has been used in the treatment of relapsed/refractory multiple myeloma. This study is to examine whether sub-lethal doses of bortezomib (Borsub) attenuate angiogenesis in a choroidal neovascularization murine model.

Methods:Laser photocoagulation was used to create choroidal neovascularization in mice. The volume of angiogenesis was assessed with isolectin GS-IB4 labeling in choroidal flat-mount. The content of vascular endothelial growth factor (VEGF) in the supernatants of ARPE-19 cell cultures was examined with the enzyme-linked immunosorbent assay (ELISA). The toxicity of menadione (VK3) and the protective effect of Borsub against VK3 were studied in ARPE-19 cells with cell survival assay. Autophagy activation in ARPE-19 cells by Borsub was examined with western blotting and immunofluorescent confocal laser microscopy. …show more content…
Tumor necrosis factor alpha (TNFα) treatment increased the content of VEGF in the supernatants of ARPE-19 cultures, which was reduced by pretreatment with Borsub. Pre-treatment with Borsub reduced toxicity from menadione in ARPE-19 cell cultures. Borsub increased LC3-II conversion from LC3-I and the co-localization between LC3 staining and lysosome in ARPE-19
