Purpose:Bortezomib has been used in the treatment of relapsed/refractory multiple myeloma. This study is to examine whether sub-lethal doses of bortezomib (Borsub) attenuate angiogenesis in a choroidal neovascularization murine model.

Methods:Laser photocoagulation was used to create choroidal neovascularization in mice. The volume of angiogenesis was assessed with isolectin GS-IB4 labeling in choroidal flat-mount. The content of vascular endothelial growth factor (VEGF) in the supernatants of ARPE-19 cell cultures was examined with the enzyme-linked immunosorbent assay (ELISA). The toxicity of menadione (VK3) and the protective effect of Borsub against VK3 were studied in ARPE-19 cells with cell survival assay. Autophagy activation in ARPE-19 cells by Borsub was examined with western blotting and immunofluorescent confocal laser microscopy.

Tumor necrosis factor alpha (TNFα) treatment increased the content of VEGF in the supernatants of ARPE-19 cultures, which was reduced by pretreatment with Borsub. Pre-treatment with Borsub reduced toxicity from menadione in ARPE-19 cell cultures. Borsub increased LC3-II conversion from LC3-I and the co-localization between LC3 staining and lysosome in ARPE-19
