Basic Chromatography Lab

Improved Essays
Introduction:
This Laboratory experiment mainly focused on the fundamental concepts of basic Chromatography. We had a goal of learning about the properties of color additives, studying their molecular structures, and their rates of solubility when added to a strip of paper and temporarily placed into a beaker of solution. When actually performing the experiment, it is relatively easy and straightforward to do as long as you do not increase the concentration of the dye or leave the wrong side of the folded strip in the beaker.

Procedure: Before starting this lab, acquire 2 chromatography strips, 2 watch glasses, two 25 milliliter graduated cylinder, two 250 milliliter flasks, 2 toothpicks, and 2 different colored dyes. First, you position

Using a ruler and a pencil, draw a faint line 15 mm from the bottom of the paper across the width of the strip. Measure 9.5 mm from the edge and place a dot on the line. This will be the starting point for the sample. On the top side of the strip, measure 20 mm from the top and fold across the width of the strip. Then, repeat those steps on your second chromatography strip. Obtain 2 different colored dye mixtures and dip a toothpick in each one and touch the colored tip on the dot on your chromatography strips. Wait for the samples to dry and repeat the procedures 2 more times, but do not increase the size of the spot, just increase the concentration by carefully dotting the same spot. While waiting for your samples to dry, pour 20 mL of the chromatography solvent into each of your flasks and cover them with a watch glass. Then, carefully place the chromatography strips into the flasks with the fold holding them up. Record your observations as the solvent travels up the paper and the chromatogram develops. When the chromatography solvent is within 1-2 cm of the fold in the chromatography strip, stop the run by removing the strip from the flask. With a pencil, lightly draw a line to mark the distance the solvent traveled.
