Abstract: A simple, novel, and sensitive method developed for estimation of Dappagliflozin and Saxagliptin. The method was carried out on reverse- phase C18 inertsil column, with mixture of Methanol: Water ( 80:20) ratio as a mobile phase, 1 ml / min flow rate. Different analytical parameters such as linearity, precision, accuracy, Limit of detection (LOD), Limit of Quantification (LOQ), robustness were determined according to the International conference on Harmonization ICH guidelines. The regression analysis for the linearity data analysis for the linear data analysis shows a good correlation coefficient of Dapagliflozin and Saxagliptin with r = 0.999 in the concentration range of 20-70 g /ml. pH meter: Weighing balance. Materials:
HPLC grade Methanol, HPLC grade analytical water, Acetonitrile, Membrane filter paper- 0.45µm was used.
Selection of Detection wavelength: Individual dilutions of Dapagliflozin and Saxagliptin were prepared from standard stock solutions using methanol. Both the solutions were scanned over the range of 200-400nm on UV-VIS spectrophotometer, using medium scan speed. From the overlay spectra, using isobestic point (which is the wavelength where the molar absorptivity is the same for two substance that are inter- convertible), 210 nm has been selected as the detection wavelength for HPLC method. This wavelength was selected and at this wavelength, the response of the drugs was adequate.
Chromatographic condition:
The mobile phase consists of mixture of methanol and water in 80:20 v/v ratio. The mobile phase filtered through the membrane filter and degasssed. The mobile phase pumped through the column at 1ml/ min flow rate. The column temperature maintained at ambient Condition. The eluents was determined at 210 nm.
