Immunocytochemistry (ICC), which this lab is based on, is commonly used as a laboratory technique to anatomically observe the location of a specific protein or antigen in cells by the use of a specific primary antibody that binds to it. A staining reagent or chromagen is used to help detect certain proteins used in this experiment, called 3,3'-Diaminobenzidine (DAB). 3,3'-Diaminobenzidine (DAB)
DAB, a derivative of benzene, is an organic compound, which is both chemically and thermodynamically stable, used in the staining of nucleic acids and proteins, most commonly for immunohistochemical procedures. Although it is water soluble in its unoxidized form, it forms a water-insoluble brown precipitate when oxidized. DAB is oxidized in the presence

EA & JB 2 Identifying Neisseria sicca INTRODUCTION Bacteria are microorganisms constituting a large domain of prokaryote. They are typically a few micrometers in length and have a number of shapes ranging from spirals to rods and spheres. Bacteria are known to be among the first forms of life to appear on Earth, and are present in almost all habitats. They inhabit water, soil, radioactive waste, acidic hot springs, and Earth’s crust deep portions. Bacteria live in symbiotic and parasitic relationships with animals and plants.…

Separate primary antibodies from different species are used along with a secondary antibody from a different species from which the primary antibody was extracted in order to create fluorescence in different parts of the cells, such as the nucleus or the mitochondrion. The purpose of Immunohistochemical staining can evaluate the presence and location of proteins in tissue sections. Immunohistochemical staining enables the observation of processes in the context of intact tissue which is very useful in the ability to assess the progression of disease such as cancer. Some disadvantages are that the procedure is less sensitive quantitatively than immunoassays, such as western blotting or…

Bromophenol Blue is a common used pH indicator dye and is with low pKa (Andrew 1995). As an acid-base indicator, bromophenol blue will lose a proton when pH of the environment around this indicator is higher than the pKa of the dye. This displacement changes the electronic distribution within the molecule and the indicator changes its color from yellow to blue (Liam, 2003). Filter paper is used to absorb bromophenol blue solution to make BPB indicator strips. After strips are exposed to the headspace of samples, bromophenol blue will react with volatile biogenic amines and change color from yellow to blue color.…

The conditions from the trypsin and biorad must remain the same throughout the experiment. Other suggestions include making dilutions from the original fractions and not from a separate test tube as that introduces new variables to the mix such as an uneven distribution of trypsin when making dilutions in a new tube. This experiment further dove into the multiple application of affinity chromatography such as examining the biological interactions of antigens, hormones and other elements. One example of this use is by Dr. Chi Zhang to purify antibodies. Antibodies have multiple uses especially in the medical field.…

Iodoform, CHI3 is formed in methyl ketone, which gives yellow precipitate when there is a positive reaction. Iodoform test is performed in basic condition as the alpha proton is eliminated in basic condition. The elimination of the alpha proton causes the alpha carbon to be nucleophilic and allows it to attack iodine molecule. The presence of the iodine in the alpha carbon makes the two other hydrogens more acidic, therefore the two hydrogens get attacked by the nucleophilic base until all of them are replaced by the iodine. Then, the bond between carbonyl carbon and the carbon containing the iodine molecules will break and form an acid salt and iodoform which is a yellow precipitate (Razote, et al.,…
