What are the 5 steps in the viral life cycle?
Adsorption
Entry into the cytoplasm
Biosynthesis of viral components
Assembly of components into complete viral units
Release from the infected cell
Is adsorption a current target of drug therapy?
No drugs presently prevent adsorption
How do immunizations work?
Immunization leads to antibodies that bind to viral attachment proteins, PREVENT ADSORPTION and prevent infection.
Name the various targets available for anti-virals and for what diseases they are used for.
Entry inhibitors - HIV
Ion Channel Blockers - Influenza
Polymerase Inhibitors - Herpes-family viruses, HIV, Hepatitis B
Protease Inhibitors - HIV
Release Inhibitors - Influenza
What drug is a Fusion Inhibitor?
Enfuvirtide
What two drugs are Ion Channel Blockers?
Amantadine
Rimantadine
How do these drugs work?
They block a proton channel on the endosome enclosed virus called M2. This prevents proton entry into the virus which prevents the uncoating of the virus and release of its genetic material into the cell.
What is the limitation of these agents?
Effective only against Influenza A
What are the adverse effects of Ion Channel Blockers?
Dopaminergic
Adrenergic
CNS side effects
What drug works as a Release Inhibitor?
Oseltamivir - Tamiflu
How does this drug work?
It is an analogue of sialic acid that inhibits neuramidase, the enzyme that cleaves sialic acid and releases the new virion. Release of new virions ceases.
Can be taken prophylactically
What are the problems with Oseltamivir?
Must be taken within 48 hours of the onset of symptoms
On average, only shortens the duration of illness by one day
Retail cost is 100$ for a typical course
