beta hemolysis = complete dissolution - Streptococcus Group E
Mannitol Salt
Selective Differential
Staphylococcus = +
S. epidermidis = negative
MacConkey Agar
red colonies = lactose fermenters -- E. aerogenes, K. pneumoniae
RED COLONIES + PRECIPITATION = E. Coli
Colorless/no lactose -- Salmonella, Shigella, Proteus
EMB
Lactose: E. Coli - blue-black colonies with green metallic sheen
E. aerogenes, K. pneumoniae both pink colonies
Nonlactose-colorless --- Salmonella, Shigella, Proteus
Triple Sugar Iron (TSI) Slant
K/NC = negative
K/A = glucose positive
A/A = lactose and/or sucrose positive
gas = cracks in agar
black precipitate = H2S positive
Lactose Fermentation
yellow = positive
AG = E. Coli, Klebsiella & Enterobacter
Negative = Salmonella, Shigella, and Proteus
Indole Production
red = positive test - E. Coli
no color - neg. test - Klebsiella and enterobacter
Methyl Red
E. Coli, Shigella, Proteus, and Salmonella = +
Voges-Proskauer (VP)
Enterobacter Aerogenes = +
Citrate Test
growth = blue slant + for E. aerogenes, K. Pneumoniae, Salmonella, Proteus, Psuedomonas aeruginosa.
Hydrogen Sulfide Production
Black precipitate = +
= Salmonella, and Shigella
Motility
E. Coli, Enterobacter, salmonella, proteus, psuedamonas aeruginosa, alcaligenes faecalis
Klebsiella & Shigella are negative
Urease Test
Deep Pink = positive -- Proteus, K. pneumonia are only positive ones
Casein
cleared section around growth meaning hydrolysis of peptide bonds has occurred = only psuedomonas aeruginosa
Nitrate Reaction
Red w/o Zinc = Positive
No color after Zinc added = positive
Only negative for this test is Alcaligenes Faecalis
Catalase Reaction
bubbles = positive test
only negative for shigella and streptococcus lactis
Oxidase Reaction
positive = pink maroon-black within 30 minutes
+ only for Psuedamonas aeruginosa and alacaligenes faecalis
Starch Hydrolysis
Added Gram's Iodine and Clear Zone around = positive test
None are positive except Bacillus cereus
Litmus Milk
lactose ferment = pink
white = reduction of litmus; excess H+ production, alkaline = blue.
Gelatin Hydrolysis
liquid = positive test. Positive = B. cereus and P. aeruginosa and proteus mirabilis
Glucose Hydrolysis
Yellow = fermentation
Positive = E. Coli, E. aerogenes, K. pneumoniae, Proteus, and Salmonella
