What cytokines activate NK cells for innate immunity?
IL-12
IFN-alpha
IFN-beta
What is a source of Il-12 to activate NK cells?
Macrophages which have ingested bacteria
Once IL-12 activates the NK cell, what can it then do?
Produces IFN-y which indirectly activates the macrophage itself to increase its metabolism to kill the bacteria.
What is the direct way that NK cells kill virus infected cells and tumor cells?
By binding cells that lack MHC, and secreting perforin and granzymes into the cells.
What is a non-chemical mediated pathway of NK cell killing?
Fas-FASL interactions
What antibody isotype is involved in ADCC using NK cells?
IgG
How does ADCC work with NK cells?
IgG coats the virus-infected cell; the Fc-gamma receptor on NK cells binds the IgG and allows the NK cell to kill it.
If NK cells don't require MHC to allow them to kill cells, what prevents them from killing normal cells in the body?
The presence of MHC class I on all nucleated cells acts as a ligand for the Inhibitory receptor on NK cells.
How do NK cells recognize virus-infected cells?
They are class I MHC negative, so the lack of the inhibitory signal tells the NK cell to go ahead and kill it.
What are intraepithelial lymphocytes?
The small population of Tcells with gamma-delta TCRs!
What is special about gammadelta Intraepithelial lymphocytes (IELs)?
They can recognize antigen in the ABSENCE of MHC.
Where are intraepithelial lymphs expressed?
At mucosal surfaces
