First bacterium shown to be the cause of a disease? (Robert Koch)
Bacillus Anthracis (Anthrax)
The thick protein coat make up of keratin surrounding an endospore is called?
Exosproium
What is an Endospore?
1.) Dormant form of bacteria.
2.) Produced when cells are stressed.
3.) Resistant to: Bacillus and Clostridium.
What does Acid Fast mean?
Resistant to decolorizing by adding alcohol after staining.
A smooth glycocalyx is referred to as a?
Capsule
What are Flagella?
Thin structures used for motility.
Name the Flagella Arrangements:
1. A single flagellum can extend from one end of the cell is said to be Monotrichous.

2. A single flagellum can extend from both ends of the cell - Amphitrichous.

3. Several flagella (tuft) can extend from one end or both ends of the cell - Lophotrichous

4. Multiple flagella may be randomly distributed over the entire bacterial cell - Peritrichous.
What is the purpose of the Quadrant Streak Plate Method?
To isolate single colonies. (Single colonies arise from a single cell)
pH Range

Acidophiles:

Neutrophiles:

Alkaliphiles:
Below pH5.5

Between 5.5 and 8.5

Above pH8.5
What happens when you place an organism outside its normal pH range?
You denature the proteins and disrupt the membranes.
What kind of stain is used for Flagella and why?
Mordant Stain is used to enhance or make larger.
What are Capsules made up of and what do they do?
Polysaccharides and polypeptides and they adhere and evade phagocytosis.
What are the Acid Fast Pathogens?

Acid Fast Genus?
Tuberculosis and Leprosy

Myco-bacterium
Ubiquity: Open Plate Tests for what?
Air contamination
Ubiquity: Closed Plates test for what?
Media Sterility
3 hand washing micro-biota
1.) Diptheriod: Gram + bacteria ie: Diptheria

2.) Staphylococci: Non-pathogenic Staphylococci Epidermidis (SE) and potentially pathogenic Staphylococci Aureus (SA)

3.) Yeasts and Fungi: Normal inhabitants of the skin
Basic Aseptic Technique
1. Clean workspace w/ vespene
2. Wash hands
3. Work near open flame
4. Always heat flame inoculation tools
5. "Briefly" heat flame open culture tube
Quad Streak Method
1. Heat flame loop and top of open tube.
2. Touch loop to bacteria (2 sec).
3. Streak bacteria on plate in "quad" formation.
4. Incubate @ 25 degree C. label side up.
