TSIA
*triple sugar iron agar
*big tube slant red in color
*differentiates member of enterobacteriaciae and other gram - rods.
TSIA results
*yellow slant/yellow butt = glucose and lactose and/or sucrose fermentation.
*red slant/yellow butt = glucose fermentation only
*red slant/red butt = no ferm.
Oxidase
*Differentiates between organisms that contain cytochrome c oxidase and those that do not.
*Indicator for Neisseria bacteria, and enterobacteriaciae(-) vs. pseudomonas(+)
Oxidase results
a dramatic color change to blue within 20 seconds indicated a positive test.
MIO
Motile or non-motile?
Indole - does the org. have tryptophanase?
Ornithine - forming alkaline by products.
MIO results
Motile = cloudy, NM - you can see the original stab line.
Indole = is there a red ring layer on top.
Ornithine - if bromocresol blue changes from yellow to purple then its (+)
Citrate
*dark green small slant tube
*Indicator is bromothymol blue
*Can org. use citrate as sole source of Carbon?
*Enterobacteriaciae & gram (-) rods
Citrate results
(+) if even small amount of blue is present or growth is present.
LIA
*lysine iron agar, big purple slant
*looks for enteric bacteria's ability to decarboxylase or deaminase lysine
*Salmonella and Shigella
LIA Results
*P/P = lysine deaminase (-) and lysine decarboxylase (+)
*P/Y = lysine deaminase (-), lysine decarboxylase (-) and glucose fermentation.
*R/Y = lysine deaminase (+), lysine decarboxylase (-) and glucose fermentation.
*black precipitate = sulfur reduction
Coagulase
*milky white color
*differentiates staphylococcus aureus from other gram (+) cocci.
Coagulase results
(+) =within 24 hours plasma has coagulated(thickened)
OF Glucose
*sealed w/ oil
*indicator = bromo blue; yellow @ 6.0 and green @ 7.1
OF Glucose results
*if sealed is green or blue and unsealed has any amount of yellow then its oxidation.
*if both sealed and unsealed are all yellow then either oxidation and fermentation or just oxidation took place.
*if both are slightly yellow at tops then oxidation and slow fermentation or just slow ferm. took place.
*If results are green or blue then no O or F took place.
MIO is a semi-solid medium
Hippurate =
*Sodium hippurate is injected into tubes, and then 5 drops of Ninhydrin are added. incubate for ~20 minutes.(+) = purple color darker than control tubes color.
