How to perform TSI
Select colony with wire, stab the butt, streak wire up the slant
Leave cap on loose incubate 35-37° C
TSI results are reported as:
Slant/butt/gas/H2S
Possible slant/butt results on TSI
A: Acid
K: Alkaline
N: Neutral

If H2S positive butt is always acid.
Positive Gas Results on TSI
Tears or bubbles in the agar
Positive H2S Results on TSI
Black color on stab line
How to perform SIM
Stab butt, leave cap on loosely, incubate 24-48 hr
SIM results are reported as:
H2S/Indole/Motility
Recorded in the same order as the initials of the test
Positive results of SIM
H2S: Black
Indole: Red color after Indole reagent
Motility: Cloudy Agar

If black is contained to stab line: NOT motile, if black spreads throughout media IS motile.
How to perform Simmons Citrate & Positive Result
Streak slant, do NOT stab, incubate
Positive: Blue
Neg: Green
How to perform Phenylalanine test
Streak the slant, do NOT stab,
Incubate 18-24 hr
Positive Phenylalanine test
After adding 1-2 drops Ferric Chloride (FeCl), immediate color change to green. No green negative.
How to perform Urea test
Mix colony into broth or streak wire, incubate overnight.
Possible Urea test results
Pink: positive
Salmon: weak positive
Yellow: negative
