What are the signs of Depression?
- loss of interest in pleasurable activity, changes in appetite, disturbances in sleep, depressed mood, loss of energy and suicidal thoughts
What are the signs of Manic depression?
increased interest in pleasurable activity, decreased sleep, increased talkatativeness, distractibility,inflated self-esteem
What is the mechanism of action of anti-depressants?
they arise from defects in neurotransmission at dopamine, serotonin and norepinephrene

1. Tricyclic compounds

What are the effects of tricyclic antidepressants?
- do not elevate mood or arousal in healthy patients (actually cause fatigue)
- rise in patient modd & interest in activities after 2-4 weeks
- potent inhibitors of norphenephrine and serotonin receptors
- downegulate beta adrogenric receptors and sensitize postsynaptic serotonergic receptors
examples: desipramine (Norpramin, imipramine (Tofranil) & notripytline (Aventyl)
What is an example of monoamine oxidase?
-iproniazid
-inhibits MAO A & B but A is more important in relieveing depression
-works after 2-3 weeks
- increases 5-HT, NE and DA but increase in 5-HT is most important

- excessive CNS stimulation
- peripheral vascular effects
- atropine effects (dry mouth, constipation, disturbances
