Amphetamine related: Amphetamine,methamphetamine, methylenedioxymethamphetamine, methylphenidate
MOA:Taken by uptake 1--by VMAT--gets into the synaptic vesicle
Effects: Increased in motor activity, euphoria & excitement, sympathetic stimulation(peripheral effect)
Good oral absorption
Medical use: mainly by methylphenidate; ADHD,by increasing dopamine. Narcolepsy, by increasing excitement
SE:anorexia,psychotic behaviour, sympathomimmetic effects
Methylenedioxymethamphetamine(ecstasy as party drug):inhibits 5HT uptake+release of 5HT SE:acute hyperthermia,water intake, heart failure
Methylxanthine derivatives: Theopylline, caffeine
Origin:coffee,tea,or cola plant
MOA: CNS--antagonism as A2 purine rec.
Peripheral--inhibition of phosphodieterase;;B agonist like effects
Effects:CNS stimulation in terms of concentration and reduction of fatigue. Doesnt cause euphoria & less locomotor stimulation. Diuresis,stimulation of cardiac muscle, relaxation of smooth muscle(bronchial muscle).
Good oral absorption.
Only theophylline is used medically
Cocaine
MOA: Inhibits uptake 1---sympatomimmetic effects and CNS effects
Effects: euphoria,increased motor activity, magnification of pleasure, enhanced mental capability.
given as nasal injection and i.v infusion
No medical use, but it is an ester type local anesthetic
