Oxidase Test
to determine if an organism produces the enzyme cytochrome oxidase
Blue/Purple=Positive
Colorless= Negative
*Must read within 30 secs to be accurate*
PDA
Phenylalanine Deaminase
(Starts out yellow)
To determine if an organism can produce PDA
Add 6-8 drops of 10% ferric chloride
Dark Green= Positive
Remains tan=Negative
MacConkey Agar
(starts out red)
Hot pink/ red= Lactose Fermenter
Colorless= Non-Lactose Fermenter
Hektone Enteric Agar
HEA
(starts out green)
Yellow/Orange= Lactose Fermenter
Blue/Green= Non-Lactose Fermenter
Black= H2S producer
Kligler's Iron Agar
KIA
(starts out orange)
To determine if an organism has the ability to ferment lactose and/or glucose, and produce hydrogen sulfide H2S
Yellow= Positive
Lactose on top
Glucose on bottom
Black= H2S
Citrate
to determine if an organism has the ability to utilize citrate as a sole carbon source
Blue= Positive
