Masson trichrome
-differentiate btwn collagen and smooth muscle
-Bouin solution= mordant; Weigert iron hematoxylin= nuclear stain; biebrich scarlet-acid fuschin= acidophilic components; phosophomolybdic /phosophotungstic acid= removes primary stain from collagen; aniline blue= collagen stain
-Collagen= blue, smooth muscle & cytoplasm= red; nuclei= black
Gomori one step trichrome
-Bouin→Weigert hem→gomori stain (combination of plasma and connective tissue stain in phoshotungstic acid)
-nuclei= black; collagen & mucus= green; cytoplasm & muscle= red
van Gieson
-van Gieson solution= picric acid + acid fuschin
-stains collagen red and muscle yellow
-used as counterstain for the Verhoeff elastic stainm
Verhoeff
-stains elastic fibers
-Verhoeff stain→sodium thiosulfate (removes excess iodine)→ferric chloride (differentiates)→van Gieson
-elastic fibers= black; nuclei= blue to black; collagen= red; other tissue elements= yellow
Gomori aldehyde fuschin
-stains elastic fibers
-aldehyde fuschin=primary stain; light green= counterstain
-elastic fibers= deep blue to purple; other tissue elements=green
Movat pentachrome
-demonstration of mucin, fibrin, elastic fibers, muscle, & collagen
-nuclei & elastic fibers= black, collagen=yellow; mucin=blue; fibrin= intense red; muscle=red
Reticulum
-ammonical silver impregnation stain that stains for reticular fibers in tissue
-reticulin stains black
Phosphotungstic acid hematoxylin (PTAH)
-demonstrates muscles striations and fibrin
-cross striations and fibrin= blue; nuclei=blue; collagen= red-brown
Periodic acid-Methamine silver
-delineates/stains glomerular basement membranes
-silver impregnation stain w/ periodic acid as the oxidizer
-basement membrane= black, background= green
Oil red O
-demonstrates neutral lipids in frozen sections
-Oil red O= primary stain; Harris hem= nuclear stain; propylene glycol=dye solvent/differentiator
-Fat= red; nuclei= blue
Sudan black B
-demonstrates neutral lipids.
-most sensitive of lipid dyes
-fat= blue-black; nuclei= red
Touidine blue
-demonstrates mast cells which stain metachromatically
-mast cells= deep violet; background= blue
