How do materials get to the lysosome?
-biosynthetic ER pathway
-Endocytic pathway
-Autophagic
-Phagocytic
How are lysosomal proteins transported from the TGN of the golgi?
- hydolases use 2 signals
1. signal 3D patch read by glycosyltranferases i cis-G that add mannose 6-phosphate

2. Mannose 6 - phosphate marker is read by receptors in the trans G network

Then the proteins are shipped out with Man-6-Phosp to the Golgi.
What ensures the acidit of a late Endosome/lysosome?
an ATP H+ pump.
Whjat is the scavenger pathway?
-Lysosomeal proteins go to the PM first then are picked up by early endosome.
What is the problem with Hurler;s?
-glycosaminoglycans (ECM) proteins which should be taken up by fibroblasts, builds up
- doesnt have the right lysosomal enzyme
-
How do you treat Hurler's?
-introduce normal cell, which will secrete some needed hydrolase that got away.
- Scavenged using PM Mannose6-P receptors
What is I-cell disease?
-No M6-P tag.
- no sorting and tragetting of huydrolases.
Why does the cell need endocytosis?
-uptake of nutrients
-clear harmful substances
-maintain surface-volume ratio
