How does reverse hplc work?

Uses silica gel reacted with hydrocarbon chains to make it non polar then a polar solvent is used instead. This means polar solvents will flow through the column first.

What is the significance of retention time?

It is the time taken for compound to flow through the column to the detector. But there are many variables so conditions must be controlled if this is used as a method of identitifing a compound. The detctor is a UV- visible spectrometer.
