Phenylephrine

WHAT? a1 selective adrenergic agonist, direct sympathomimetic


MOA? Activation of PLC/ Inositol pathway. Leading to release of intracellular Ca2+.


EFFECTS? All smooth muscle excpet that in the G.I contract in responce to stimulation of a-1 receptors due to Ca2+ release.Large arteries and veins and also arterioles are constricted, decreased vascular compliance, increased peripheral resistance (increased BP)a-1 receptors stimulate smooth muscle proiferaction in blood vessels and prostate. This is important in benigh prostatic hyperplasia (treated with a-1 antagonists)


USES? Nasal decongestant


Dobutamine

WHAT? B1 selective agonist, sympathomimetic.


MOA? Agonises B1 adrenoceptors in the heart, increase in HR and contraction force


USES? Cardiac failure

Clonidine

WHAT? Selective a2 adrenoceptor agonist, direct sympathomimetic


MOA? Decrease in Adenyly cyclase activation, decrease in cAMP and Ca2+ channel activation and phosphorylation. Inhibitory effect


EFFECTS? Nerve terminals:Presynpaptic adrenoreceptors present on both cholinergic and NA nerve terminals. Inhibitory action due to a-2 actionMetabolism: Inhibition of insulin secretion due to a-2, leads to adrenaline-induced hyperglyceamia


USES?

Salbutamol, talbutaline, sulmeterol

WHAT? B2 selective agonist, sympathomimetic


MOA?Agonises B2 adrenoceptors on smooth muscle, increase of cAMP leafs to increase of Ca2+ extrusion and reuptake.


EFFECT? Relaxation of smooth muscle and vasodilation of vascular beds.


USES? Asthma (reversible obstructive pulmonary disorders)
