Class: TH-inhibitor
Action: competitive inhibitor of Tyr for TH, ↓NE, epi syn.

Notes: Can cross BBB; active in CNS.
reserpine
Class: vesicular uptake inhibitor
Action: irreversibly inhibits uptake pumps; new vesicles with new pumps must be made to compensate. Depletes NE, which are exposed to MAO outside of vesicles.
Tx: anti-HTN
Side effects: crosses BBB, has CNS side effects. May cause cardiac arrhythmia. Rarely used today.
Notes: At high doses, MAO may be overwhelmed by sudden release of NE, creating transient HTN.
guanethidine
Class: vesicular release inhibitor
Action: depletes vesicular NE; inhibits release of endogeneous NE.
Tx: anti-HTN, rarely used today
Side effects:
Notes: Must be taken up by reuptake pump to exert effect. Can't cross BB, highly polar. All effects peripheral.
cocaine
Class: reuptake inhibitor
Action: inhibits reuptake of DA in CNS, NE in periphery, but blocks effect of tyramine.
Tx: anti-depressant
Side effects: in the periphery, can cause excessive sympathomimetic effects on cardiovascular sys.
Notes: Sympathomimetic effect of tyramine paradoxically ↓'d by cocaine.
amphetamine
Class: indirect sympathomimetic; monoamine reputake inhibitor
Action: binds to monoamine transporters, ↑g NE, DA in cleft. Blocks reputake of NE, DA. Sxr is similar to NE, but resistant to MAO/COMT.
Tx: ADHD
Side effects: HTN, tachyarrythmia.
Contraindications: glaucoma, MAOIs, HTN, hyperthyroidism.
Notes:
ephedrine
Class: mixed-acting sympathomimetics
Action: NE agonist that resists MAO/COMT degradation
Tx: nasal decongestant, brochodilator, pressor agent.
Side effects: HTN, tachycardia.
Notes:
α-methyldopa
Class: α2-agonist (centrally-active), DA-analog, general α-agonist in periphery.
Action: a prodrug; crosses BBB and met.'d into α-methy-NE.
Tx: anti-HTN
Side effects: urinary retention; often given with a diuretic.
Notes:
phenylephrine
(Sudafed PE, Neo-Synephrine)
Class: α1-agonist
Action:
Tx: nasal decongestant, hypotension
Side effects: cardiac arrhythmia, HTN.
Contrain.: glaucoma, HTN, tachycardia.
Notes: replaces pseudoephedrine due to use as methamphetamine ingredient.

Action: blocks CNS presynaptic α2-R, ↓ NE release.

Notes: Does not block systolic HTN effect of Epi.
prazosin
Class:α1-antagonist
Action: ↓peripheral BP without ↑g HR (β1 effect).
Tx: anti-HTN
Side Effects:
Notes: usu. used in conjunction with other classes.
metoprolol
Class:β1-antagonist
Action:
Tx: HTN, performance anxiety, wide-angle glaucoma.
Side Effects:
Notes: will only affect heart, unlike nonspecific β-blockers.
carvedilol
Class: nonselective adrenergic antagonist
Action: blocks α1, β1, and β2 RR. Theraputic effect poorly-understood.
Tx: CHF, HTN
Side Effects:
imipramine
Class: reuptake inhibitor
Action: inhibits NE reputake.
Tx:
Side Effects:
epinephrine
Class: nonselective adrenergic agonist
Action: ↑HR (β1), systolic pressure (α1), and C.O., ↓ some peripheral vascular resistance.
Tx: anaphylactic shock, cardiac arrest.
Side Effects: cardiac arrhythmias.
Contraindications: β-blockers, which will cause severe HTN and cerebral haemorrhage. Narrow angle glaucoma.

Action: dilates renal BVs
Tx: compromised renal fxn, shock.
