Preparation of working standard solution in 0.1N Hydrochloric acid: Prepared 1 µg/ml, 2 µg/ml, 3 µg/ml, 4 µg/ml, 5 µg/ml, 6 µg/ml, 7 µg/ml, 8 µg/ml, 9 µg/ml, 10 µg/ml solution, take to from 100 µg/ml respectively 0.1ml, 0.2 ml, 0.3 ml, 0.4 ml, 0.5 ml, 0.6 ml, 0.7 ml, 0.8 ml, 0.9 ml, 1.0 ml than make up mark of 10ml volumetric flask with 0.1N HCl. The absorbances of the solutions were measured against 0.1N HCl as blank at 274 nm using UV spectrophotometer. The absorbance values were plotted against concentration (µg/ml) to obtain the standard calibration curve.
