Phenylalanine Deaminase Test Lab Report

Decent Essays
The possibilities of two unknown microorganisms lead to phenylalanine deaminase test. The detection of phenylalanine deaminase enzyme was carried out by adding ferric chloride to phenyaline slant. When ferric chloride was added to phenylalanine deaminase tube, it turned into army green color( positive result) which indicated that unknown microorganism contains phenylalanine deaminase enzyme. On the other hand, no color change in the slant indicates negative result for this test. The choices between two…………………. was narrowed down By performing this test. The positive result for phenylalanine deaminase indicated the unknown microorganism #404 is P.vulgaris.

To check motility of an unknown microorganism, motility test was performed. For motility

Reduction involves increasing the number of carbon hydrogen bonds by adding hydrogen across a double or triple bond which results in an increase in electron density at the carbon atom. Organic functional groups containing double and triple bonds which undergo reduction are unsaturated. The reduction of the double bond by addition of hydrogen atoms results in the product being fully saturated. Metal hydride reducing agents that have different reactivities toward specific functional groups are commonly used in chemical reductions.…

Following Gram staining and microscopic examination, Unknown 15 was identified to be a Gram positive coccoid. Based on these results, all of the Gram negative genera were eliminated including: Enterobacter, Citrobacter, Klebsiella, Escherichia, Pseudomonas, Serratia, Alcaligenes, Neisseria, Proteus, Salmonella, Shigella, Erwinia, Veillonella, and Flavobacterium (5). Bacillus, Lactobacillus, Listeria, Kurthia, and Arthrobacter were eliminated because they did not match the coccoid shape of Unknown 15 (5). The catalase test was the next designated test in the identification of Unknown 15. Unknown 15 tested negative for the enzyme catalase as it did not form gas bubbles after the addition of hydrogen peroxide to the sample.…

After, then other biochemical tests were performed that were specific to the gram- negative bacteria to start the identification process of the unknown. The results of the gram negative tests lead to the identification of the bacteria Alcaligenes faecalis. The gram stain procedure was the first test performed by isolating a colony from the pure culture streak plate. The gram stain was followed orderly right from the referenced laboratory manual. The test resulted to be Gram- Negative bacteria from the compound microscope which were pink and rod shaped that can be seen with the 100x objective lens with the oil immersion.…

    Phage Lab Report

The first step in phage infection is the attachment of the phage to the host cell surface. This is typically accomplished by the recognition of a receptor on the outside of the bacterial cell wall such as an antigen, pilus or other structure. There is much variability from phage to phage in terms of which receptor they bind to. Bacteriophage can generally be classified into two categories, lysogenic and lytic (virulent) (Fig 6). The choice between the lytic and lysogenic cycle depends on the relative expression rates of phage repressor encoded by a cII gene (promoting lysogeny) and cro protein, capable of turning off repressor gene expression and starting the lytic pathway (Campbell, 1967).…

During the phase and process of the bacterial unknown project, vast amounts of test were performed in order to determine an unknown species of bacteria. Within the many procedures of identifying the unknown bacteria, I was first given a tube with the unknown bacteria, from which later on I started subcultures by streaking bacteria from the original tube onto a tryptic soy agar (TSA) slant. Throughout the process of identifying the unknown, new subcultures were produced in order to use the newest bacteria to conduct the tests. Thus, this process allowed the unknown bacteria to grow and maintain. Before a series of test were conducted, first a gram stain test was performed in order to determine the unknown’s gram reaction and its cell morphology.…

The study of microbiology requires not only understanding the microscopic organisms, but also the understanding of lab techniques and procedures used to identify, control, and manipulate microorganisms. The identification of microorganisms is not only important in microbiology lab, but also in the medical field to identify an agent of a disease that will help treat the patient by using the correct antibiotics to kill off the host. In this unknown lab report, techniques and procedures learned in the microbiology laboratory during the semester that was performed to test ones practical understanding of microbiology. The sole purpose of the unknown lab is to demonstrate understanding of the experimental methods and lab techniques learned during…

Using aseptic technique and using the streaking plate method. We also inoculated the unknown sample with the cotton swab onto a TSA slant, in which we transfer the unknown organism to the bottom of the slant and through a zigzag motion to make sure the TSA slant was completely covered with our unknown organism. Once the TSA plate and slant was inoculated with our unknown organism it was allowed to be incubated at 35 degrees Celsius for 24 hours. After the plates had undergone incubation for the full 24 hour duration, color and morphology was observed and noted for our unknown organism. Gram staining was than conducted after, in which we determine if the unknown organisms was either positive or negative based on the thickness of its peptidoglycan wall that was able to retain the crystal violet color or not.…

Based on the laboratory tests provided, it can be determined that the unknown organism can be identified as Pseudomonas aeruginosa (P. aeruginosa). This organism is an opportunistic pathogen that can infect non-mammalian, plants, and humans. P. aeruginosa belongs to the bacterial family Pseudomonadaceae, and is a gram-negative rod that approximately measures 0.5 to 0.8 µm by 1.5 to 3.0 µm. It can normally be found in water and soil. Its single polar flagellum makes it motile, and it is known to be one of the fastest swimming bacteria found in hay infusion and pond water (Crittenden, Trussell, Hand, and Howe 2012).…

Initially when unknown microbe #7 was gram stained and observed under the microscope, the color appeared more pink than purple, leading to the conclusion that unknown microbe #7 was gram negative. However, when completing the KOH lysis test, the microbe didn’t become sticky and remained tacky which is how a gram positive KOH test would result. It was decided to double check the KOH lysis test and the second test still yielded the same results as the first, gram positive. The conclusion of unknown microbe #7 being gram negative from the gram stain was an error.…

The experiment conducted was to observe the enzyme catalysis of Horseradish Peroxidase with different determining factors. In this experiment the horseradish peroxidase enzyme should have an optimal temperature of 35°C, should have an optimal pH of 6, and should catalyze rapidly with the substrate and enzyme concentration. We were able to observe that the enzyme reacts best with its optimal temperature of 35° C and optimal pH of 6. Moreover, when there’s a high the substrate concentration and a high enzyme concentration we can observe a rapid enzyme catalysis. The hypothesis was partially falsified for the optimal pH and the optimal temperature were both different than that stated in the hypothesis.…
