Discovering Microbes In A Mini Ecosystem
Introduction:
Every ecological environment provides with distinct nutrients that allows certain microorganism to survive and grow. Sergei Winogradsky, a Russian microbiologist, developed the Winogradsky Column to grow and investigate microorganisms in a complex biofilm communities. This column will grow different microbes at different gradients that will appear on the column. Depending on the various concentrations of nutrients, choice of soil and water used, a variety of organism will appear over time in the column (Esteban et al., 2015). This lab was discovery science. The control and experimental column showed that the black spots were appearing more where the sun light did not hit it. The control column presented small amount of green spots where the sunlight was hitting the column. Meanwhile, the experimental presented less green bacteria. The black spots increased in both columns, but the experimental group had a greater area of black spots. Finally, the experimental group had some orange spots which means that the column had non-sulfur bacteria, and above the orange layer, there was a big green spot of green non-sulfur bacteria look figure 1.3 (Urmeneta & Duró, 2011). Black spots were found at the bottom and where the sunlight did not reach the columns. The experimental column has a greater amount of bacteria that can reduce sulfur into hydrogen sulfide. Rundell et al. (2014) explained that “Deltaproteobacteria have been identified as an abundant member of black layer communities in wetlands”. As a result of high concentrations of sulfide, the column also presented large amount of green sulfur bacteria. In the experimental group, the column did not have purple sulfur bacteria. However, the experimental group showed large amount of orange spots that the control group didn’t have. Orange spots indicated that there was the presence of non-sulfur bacteria.     With this test, another test to figure out the “oxygen requirements” of the organism was done in a “thioglycollate agar tube” from which will allow the organism to grow either towards the top of the agar where oxygen exists, an “aerobic” microbe, or to the bottom of the tube where its considered “anaerobic”, or dispersed between the tube to show that it’s a “facultative anaerobe” (MB 352 Laboratory manual pg 47).
