Applying all methods learned in the microbiology laboratory class, identification of an unknown bacterium study was conducted. The purpose of the experiments is to identify what bacteria is given, and know which the name of the gram negative and gram positive bacteria. To determine the name, all methods that have been learned during the microbiology lab classes were used to determine the names.
Identifying the names of the gram positive and gram negative, (1) will give an idea about the bacteria, (2) the effect of it, (3) proper treatment will be determine in other to kill or cure these bacteria if a patient is been affected by it. (4) The bacteria might be a severely harmful or mildly harmful.

For this experiment, unknown sample #12 was used. The first step in identifying any unknown bacterium is to grow a culture on a nutrient agar plate. This bacterium was also plated on a MAC plate, which is both selective for gram-negative bacteria and differential as well. On the NA plate, bright red colonies were present; on the MAC plate there was bacterial growth as well, but no color change was observed. This lack of color change on the MAC plate indicated that the bacteria did not ferment lactose.…

A total of four microorganisms were isolated from the sample brought back from Mars. To identify the microorganisms which were isolated from the sample brought back from Mars, an rDNA sequence analysis was required. Every sequence had an S_ab score of 1.000 meaning the sequence is identical to the microorganism that it was matched to. Before gathering phenotypic results, we gathered phylogenetic data for the four microorganisms which includes Micrococcus luteus, Citrobacter freundii, Hafnia alvei, and Staphylococcus epidermidis. Micrococcus luteus is a gram positive, coccus shaped microorganism that is nonmotile and unable to grow on glucose minimal media or simmons citrate agar (Kocur, 1972;).…

The study of microbiology requires not only understanding the microscopic organisms, but also the understanding of lab techniques and procedures used to identify, control, and manipulate microorganisms. The identification of microorganisms is not only important in microbiology lab, but also in the medical field to identify an agent of a disease that will help treat the patient by using the correct antibiotics to kill off the host. In this unknown lab report, techniques and procedures learned in the microbiology laboratory during the semester that was performed to test ones practical understanding of microbiology. The sole purpose of the unknown lab is to demonstrate understanding of the experimental methods and lab techniques learned during…

This was allowed to be incubated for 24 hours at 35 degrees Celsius. The observation of color change and gas formation is what ultimately led to the identification of our unknown organism. Results: Dichotomous key Gram Stain: After performing the gram stain, our unknown organism appeared to be pink indicating that it is gram negative and that the morphology is rod shape or bacillus.…

Bergey’s manual was used to identify the unknown to be either from genus Flavimonas also known as Pseudomonas or Frateuria based on the results of the experiments. Multiple different experiments were conducted to identify the unknown, however only seven characteristics of the unknown were discovered from the data in Bergey’s manual. Not all test performed in lab were used due to the fact that some tests were not listed on the manual, while others did not apply to the category of the bacteria. Test used to find the morphology characteristics of the unknown yielded that the colony of the unknown was yellow pigmented and the shape of the cell was rod. More information was found by examine the cell wall structure of the bacteria, Gram test revealed…

When it derives to contemplating out an unknown bacterial species the whole purpose is to benefit us in order to better understand what this species is capable of. Throughout a general basis of identifying an unknown species we must figure out either it will become a gram-positive or gram-negative type of bacteria and in doing so we must do a gram staining method. This method works because it will direct us to what test we can do next based off either it is negative or positive bacteria, which is exactly what I had done for my unknown. These experiments are not just done without a reason; on the contrary, it is very important to identify these unknown bacterial organisms. For instance, in a medical point of view when becoming very sick to the…

When determining the gram reaction of the unknown, a Gram stain was done. This process differentiates between a Gram-positive and Gram-negative microorganism. This is done in four steps. The microorganism was stained with iodine, crystal violet, alcohol and acetone, and safranin.…

Introduction Identifying unknown bacterium is not only vital to microbiology, but also to the medical and pharmaceutical industries in order to treat disease/infection properly. Bacteria are classified and identified to distinguish among strains and to group them by criteria of interest (1). There are several different procedures that can be utilized to identify a bacterium. This includes the oxidation test, gram stain, and catalase test. The purpose of this lab report is to show the understanding of all procedures used to identify the bacterium, and explain how the given results led to the identification of the unknown bacterium.…

After obtaining my unknown organism the first thing I needed to do to get started was to do a gram stain. This would help me decide which kind of further testing would need to be done in order to help identify the organism that is causing gastrointestinal distress in Brown County. After doing a gram stain test on my unknown, I found that it was a gram negative organism. This prompted me to do some tests that are specifically for gram negative organisms. When I looked under the microscope I seen some rod shaped bacteria.…

The properties of selective and differential media suggest that differences between the bacterial species could be used in this lab to identify these six species but their effectiveness is unknown. The efficacy of identifying a completely unidentified bacterium in a cost-sensitive manner using these test also has yet to be determined. We hypothesized that applying a combination of selective and differential media with biochemical tests would provide a presumptive identification of the first group of six unknown bacteria. We also hypothesized the identification an unknown member of the Enterobacteriaceae family referred to as unknown #53 could be definitively identified using the API 20E and other tests. To test the first hypothesis, samples of four of the six unknown bacteria were streaked onto plates of Mannitol Salt agar, MacConkey agar, and Eosin Methylene Blue agar to examine their growth on the media.…
