K. Pneumonia Report

After receiving unknown organism #14, many tests were performed to discover the identity. After running a variety of tests, my results led me to believe that my unknown organism was Klebsiella pneumoniae. K. pneumonia produced a negative-rod gram stain. My organism, K. pneumoniae, tested positive for the three main carbohydrates, glucose, lactose, and sucrose. After running tests on these carbohydrates, a pH indicator color change from red to yellow signified a positive result, while remaining red, signified a negative result (Lab). All three tubes turned yellow, which told me that K. pneumonia ferments glucose (w/ average gas), lactose (w/ less gas), and sucrose (w/ lots of gas). Urea hydrolysis was another test performed with a positive outcome. Being that there was growth on my media, this determined that K. pneumonia can utilize urea for growth in replacement of energy and oxygen (Urea). After performing the methyl red test, a negative result was obtained because it remained red. This negative result implies that my organism does not perform mixed acid fermentation with glucose (Acharya, ref #3). The Voges-Proskauer test gave a positive result. This positive result is “indicating the presence of diacetyl, the oxidation product of acetoin (Acharya, ref #2). The last test performed was the nitrate reduction test, which determined if the organism could reduce nitrate to nitrate via nitrate reductase enzyme (Welcome, ref #2). After this test was completed, I got a positive result. K. pneumoniae can grow on any ordinary nutrient media. When introduced to solid media, will form protruding, shiny, mucous, circles (Klebsiella). K. pneumoniae is a facultative anaerobe (Klebsiella) with a temperature preference of thirty-seven degrees celsius, making humans the perfect home to these great

    My professor had suggested that instead of me continuing on ahead doing the catalase test she had recommended that I do a mannitol salt agar test or a Blood agar test for my final test, which would help determine between Streptococcus pneumoniae, Staphylococcus epidermidis. I decided to go along with the mannitol salt agar method and again with my inoculated loop I grabbed some of my unknown #15 and I had applied it to aseptic technique to the agar plate.
