The coagulase test is used to isolate Staphylococcus to clot plasma. There are two types of coagulase enzymes: bound and extracellular. The bound enzyme is attached to the cell wall of Staphylococcus aureus and is detected on a slide. The extracellular coagulase is secreted by S. aureus into the environment and detected in a tube. The coagulase enzyme converts fibrinogen to fibrin which results in a clot. It is read every 30 minutes for up to 4 hours to determine if agglutination occurred and an ID of Staphylococcus is possible.
This test will detect if an organism contains the hippurate hydrolysis or hippuricase, which breaks down sodium hippurate into benzoate and glycine. This test will identify Group B Streptococcus by turning a deep purple/blue-ish
