After obtaining my unknown organism the first thing I needed to do to get started was to do a gram stain. This would help me decide which kind of further testing would need to be done in order to help identify the organism that is causing gastrointestinal distress in Brown County. After doing a gram stain test on my unknown, I found that it was a gram negative organism. This prompted me to do some tests that are specifically for gram negative organisms. When I looked under the microscope I seen some rod shaped bacteria. So what is this organism and is it causing the reported cases of GI distress? The gram stain I did showed a pink organism which told me that it was gram negative. I then continued with some tests that were specifically for gram negative bacteria. I did an OF glucose test, a Citrate test, SIM test which told me the H2S, Indole and motility results and also a urease test. After letting my tests incubate I found that there were negative results for the H2S, Indole and urease tests and positive …show more content…
The symptoms reported were vomiting, bloody diarrhea, and severe cramping which is a gastrointestinal infection. It was also mentioned that raw bean sprouts were possibly the culprit for this and people were warned not to consume them. Although it can cause GI distress in some as mentioned earlier, Enterobacter aerogenes is a hospital acquired infection. This would mean that all the cases that were reported would have had to have been associated with a recent hospital stay. The main ways to acquire this in the hospital is when you are in the hospital a long period of time, have a catheter, have a surgical procedure or even antibiotic use in the past (bioquell). Because of the fact that the reports don`t specify that the cases were from eating raw bean sprouts while staying in the hospital tells me that this isn`t the disease that caused the outbreak in Brown

By comparative analysis of biochemical tests that were performed throughout the semester to find out its characteristics and determine the identification of bacterium out of four potential unknown was given. To figure out the characteristics, T-streak was performed to isolate the sample from broth to a Tryptic Soy Agar plate, along with an additional slant in case something wrong with the original plate that can possibly affect the result.…

After, then other biochemical tests were performed that were specific to the gram- negative bacteria to start the identification process of the unknown. The results of the gram negative tests lead to the identification of the bacteria Alcaligenes faecalis. The gram stain procedure was the first test performed by isolating a colony from the pure culture streak plate. The gram stain was followed orderly right from the referenced laboratory manual. The test resulted to be Gram- Negative bacteria from the compound microscope which were pink and rod shaped that can be seen with the 100x objective lens with the oil immersion.…

The Gram positive unknown was identified to be Listeria Monocytogenes. This conclusion was reached with several bits of evidence. Firstly, the Gram positive unknown was rod shape. This narrowed it down to seven different species. However, I felt it was important to perform a series of tests in order to confidently conclude that the Gram positive unknown was indeed Listeria Monocytogenes.…

When it derives to contemplating out an unknown bacterial species the whole purpose is to benefit us in order to better understand what this species is capable of. Throughout a general basis of identifying an unknown species we must figure out either it will become a gram-positive or gram-negative type of bacteria and in doing so we must do a gram staining method. This method works because it will direct us to what test we can do next based off either it is negative or positive bacteria, which is exactly what I had done for my unknown. These experiments are not just done without a reason; on the contrary, it is very important to identify these unknown bacterial organisms. For instance, in a medical point of view when becoming very sick to the…

Introduction Identifying unknown bacterium is not only vital to microbiology, but also to the medical and pharmaceutical industries in order to treat disease/infection properly. Bacteria are classified and identified to distinguish among strains and to group them by criteria of interest (1). There are several different procedures that can be utilized to identify a bacterium. This includes the oxidation test, gram stain, and catalase test. The purpose of this lab report is to show the understanding of all procedures used to identify the bacterium, and explain how the given results led to the identification of the unknown bacterium.…

Then there is a process of staining the slide first with violet, then with decolorizer, then with safranin. After the precise and delicate process of gram stain, a microscope will be utilized to view the color and morphology of the stained organism present. Viewing this slide, it is found that the organism present turned out purple, making it gram positive. This is meaningful because gram positive tells us that this organism has a thick peptidoglycan and teichoic acid cell wall. The morphology turned out to be bacillus, meaning that it is rod…

To determine the unknown’s morphology is the first step in narrowing down possible organisms. A MacConkey and Mannitol Salt Agar tests were conducted on the organism and revealed the following. No growth on the MacConkey plate resulting in a gram positive bacteria and the Mannitol Salt Agar result showed it could be Staphylococcus or Micrococcus. (1) A gram stain test was used in determining the shape, size and arrangement of the unknown bacteria (1). Identification of unknown organisms is useful in a wide variety of settings; from clinical diagnostics to environmental monitoring (1). Species of bacteria can differ fundamentally in aspects ranging from morphology to metabolism. Exploiting these differences can allow for identification of unknown organisms. Gross morphological characterizations are crude, and better replaced by more accurate and specific tests. At a fundamental level, tests such as the Gram stain divide bacteria in to two broad categories based on the characteristics of their cell wall (2).…
