Citrate Lab Report

Citrate: The citrate test gave a positive result, showing a dark blue color in the tube, caused by the alkaline conditions present, which is caused by the bacteria’s ability to utilize the citric acid present in the citrate slant. The positive result confirms the presecnce of the citrase enzyme. H2S Production: The H2S test yielded a positive result, giving rise to a black precipitate created by iron and H2S combining. Proving the presence of the desulfurase enzyme. Indole: The indole test yielded a negative result, giving a yellow ring at the top of the test tube caused by lacking a reaction between Kovac’s reagent and indole, due to the fact that there is no indole present in the tube, as well as no tryptophanase enzyme. MR test tube inoculated and incubated for 24 hours, ten to fifteen drops of methyl red indicator then added to the cultured. If it turned red then that was the positive for the presence of mixed acids. No color change was negative, however, if the media turned orange, it was inconclusive. VP test tube was inoculate and incubated up to 3 days, vortexed, added Barrits Reagent A (alpha-napthol) and Barrits Reagent B (potassium hydroxide). Red or pink color was positive reaction, no color change was negative, however it could take up to an hour to see the color change.…

To begin Lab 10 of Chem 115, students received permission to start the lab procedure after completion of prelab. First, students made a 25.0 mL of a 0.15 M Malonic Acid and 0.20 M Manganese Sulfate Monohydrate solution and labeled it "Solution 1". After the solution was made, a piece of parafilm was used to seal the flask before mixing it well. Next, a 25.0 mL of a 0.20 M Potassium Iodate solution, using 0.080 M sulfuric acid as a solvent, was made. Before adding the acid, students heated about 25 mL of the acid in a 100 mL volumetric flask.…

Assignment #2: The Replication of DNA in Escherichia Coli Due Date: Feb 8, 2016 at 9:00 AM CST 1a. A procedure in which a small amount of DNA material is immersed in concentrated solution, cesium chloride (CsCl), and soon after placed in centrifuge until the point where equilibrium is achieved is called density-gradient centrifugation. Contrasting techniques involving diffusion and sedimentation form a persisting CsCl concentration gradient. Furthermore, the concentration gradient and pressure of centrifugal force results in increased density.…

The next test, Bacitracin Susceptibility, was the next test on key that helped lead to the identification of the unknown. For this procedure, you grab a Blood Agar plate, and then streak the plate with a flamed loop containing your unknown. After the plate is streaked, you will then ask your instructor to place antibacterial discs throughout the plate. These antibacterial discs, after growth, will show the zone of inhibition which will then lead to your results. You leave the plates for a couple days in order to later on see some growth.…

Throughout the semester in the microbiology lab we have been taught how to conduct multiple experiments to identify microorganisms. Many of these experiments can be used on all different types of microorganisms but cumulatively they can help isolate a singular microorganism’s identity. At the beginning of the semester we were divided into several groups and were assigned an unknown of which we were tasked with identifying. Being assigned number 14 as our unknown, my group consisting of Chijindu Eke, Austin Dicus and myself conducted these experiments over the course of the semester and have successfully identified our microorganism to be Enterobacter Aerogenes. The plan to identify the microorganism was to grow a pure and uncontaminated…

Finally, the bacteria was tested for the oxidase test. Oxidase test looks for the presence of an enzyme that allows oxygen to be the terminal electron acceptor in the bacteria’s electron transport chain. The test has an artificial electron acceptor with an indicator that is made to change color from yellow to blue when it is in the presence of oxidase (Brown and Smith pg. 85). The bacteria was tested and produced a blue color on the swab proving it was positive for containing the oxidase…

A positive result is positive growth of cells within it, and it shows that the bacteria inoculated within it is aerobic and growth is supported. A negative result is lack of growth and it indicates that the bacteria inoculated within it is not aerobic. The test works by being selective and only allowing the growth of aerobic bacteria. SF broth is a selective medium and is used to select certain organisms by providing the correct environment and nutrients needed to grow. A positive result will elicit a color change of the media from purple to yellow.…

After the incubation period, the agar containing the antibiotic testing was taken out and if the microbe reacted with the antibiotic, a circle or clearing would appear around the disk containing the antibiotic. The diameter of this circle was then measured and compared with a table found on page 95 of the Microbiology Lab Manual to understand the antibiotics resistance, intermediate and sensitive levels, as they differ for each antibiotic. The numbers on the table in the Microbiology Lab Manual represented whether the microbe was resistant, shown in the light blue color on the graph, intermediate, shown in purple, or sensitive to the antibiotic, shown in the periwinkle color. As shown in figure 1 above, indicated with a yellow line, each antibiotic tested on unknown microbe #7 turned out to be sensitive. The diameters of the cleared out areas were vancomycin: 21mm, erythromycin:…

The result indicated that the unknown #63 was positive for lactose fermentation and eliminated S. marcescens, P. mirabilis, and P. vulgaris, which are lactose nonfermenters. BCP (lactose) broth which was originally a purple colored medium turned into a color yellow and signs of gas production was also noted. The result indicated that the unknown #63 was positive for both acid and gas production and eliminated K. pneumoniae, and E. aerogenes because they can produce acid, but no gas production was suggested from the dichotomous key. The test result for SIM deep slant that was used to test for sulfur-reducing turned up positive due to its color changed into a blackening color. The test result eliminated P. aeruginosa and identified unknown #63 as being Citrobacter freundii.…
