Introduction : Enterotube II is used to identify multiple tests in one tube. twelve different conventional media plus a self-contained inoculating wire. The twelve test are for glucose and gas, then lysine, ornithine, H2S and indole, adonitol, lactose, arabinose, sorbitol, dulcitol, phenylalanine (amino acid), urea, and citrate (No VP test require). These medias performs 15 different biochemical tests that will determine the genus and the species of gram negative bacteria. This test will help identify the 5digit number that will distinguish the organism using 5-digit interpretation code book. Enterotube has two sides blue and white. The blue side is where the organisms are inoculated and from the white side, you break the notch to puncture …show more content…
Method/procedure
1. I made sure that the table was very neatly clean
2. I labeled the the Enterotube with my name and the unknown number that was assign by my professor.
3. I opened the Enterotube from the blue end which has the handle and pull the whole needle out make sure no to contaminate it.
4. Then I scraped the sample from the assigned tube and inoculated on to the needle.
5. Inserted it back to the tube and moved it back and fourth 5 times until the agar are well inoculated.
6. The needle should be coming out all way out so, I broke the notch on the needle by bending it 2 or 3 times.
7. After breaking the notch the needle will go back in the Enterotube.
8. On the front side of the tube there are some spots called the windows. Some of these are aerobic chambers and some are the anaerobic chambers. In the aerobic chambers I poked the notch that I just broke from the needle and immediately puncture the windows.
9. Made sure to no touch it because it will contaminate while doing this.
10. After, I closed the blue end and the white end and incubated it in 35 – 37 centigrade for 18 to 24 hours with the Enterotube line in upright
