One possible limitation for this experiment was the contamination of the chromatography paper by foreign amino acids. Any amino acids that our gloves came in contact with throughout the lab transfered over to the chromatography paper and appeared as purple spots, mostly on the border of it.

Conclusion-

The amino acids in unknown mixture A were Glycine and Valine. The amino acids in unknown mixture B were Glycine and Leucine. The amino acids in unknown mixture C were Lysine and Prolise.

Experiment 11: Peptides and Amino Acids The purpose of this experiment was to study the amino acid composition of artificial sweeteners and then observe simple acid and base catalyzed reactions of the amino acid and artificial sweetener aspartame. This was achieved by performing a TLC analysis using ninhydrin as an indicator to observe the composition of the amino acid as well as two of the components that make up aspartame. This is followed by the addition of an acid or base to aliquots of diet sodas to examine the resulting reactions and compare them to a known hydrolyzed aspartame sample. The most common method of synthesizing amides is through the reaction of a primary or secondary amine with a carboxylic acid derivative.…

To make a 12.5% resolving gel, a mixture of 30% acrylamide, 0.8% bisacrylamide, 4X Tris HCl buffer with pH 8.8, DI water, 10% ammonium persulfate, and TEMED. The solution was poured into a gel caster, ethanol was added on top, and it was left alone to polymerize. A 4.5% SDS stacking gel was then prepared by mixing: 30% acrylamide, 0.8% bisacrylamide, 4X Tris HCl buffer with SDS pH 6.8, DI water, 0.25% bromophenol blue, 10% ammonium persulfate, and TEMED. The gel was left to polymerize and the running buffer was prepared similar to the previous step. Following this the protein samples were prepared by calculating the volume of protein needed to add to each sample, by taking the amount of protein given and dividing by the protein concentration found on the table completed in experiment seven (Table 1).…

Biochemical Unknown I. Introduction Cultural Characteristics or morphology and biochemical tests can be used to identify and classify microorganism. By culturing microorganism in nutrient broth, slants, and on nutrient agar plates, the cultural characteristics or morphology can be determined. In this lab, the test tube 2 was incubated in a nutrient broth. The pigmentation of the tube was yellow. The media for test tube 2 was turbid.…

Experiment Testing Organic Molecules in Commercially Available Protein Supplements Introduction In this experiment, my lab partners and I examined three different samples of protein supplements in order to test for organic molecules such as sugar, starch, and protein. The average consumer that purchases protein supplements, such as those we examined in lab, use them for physical fitness. This means that they would be searching for the protein powder with the most amount of protein and least amounts of other ingredients such as sugar and starch; however, this can be difficult to judge just from the nutrition label. Because we were interested in finding the amounts of sugar, proteins, and starch in each sample, we ran both a qualitative and…

Polarity is a characteristic of molecules that making the charges unequally distributed causing it to create a magnetic field. One way you can check if a substance is polar or not is by doing the High Performance Chromatography Lab. Using water, different percentages (5%, 20%, and 60%) of methanol and isopropyl alcohol, Kool-Aid, and a syringe with a C-18 cartridge attached. Putting the Kool-Aid through first, made the dye stay in the cartridge, and the liquid turned clear. Next, putting the methanol alcohol, from lowest to highest percentage, made the solution turn a shade of yellow, pink, and blue.…

The goal of this experiment was to determine the Michaelis constant (Km) and also the maximal velocity (Vmax) and the inhibition of alkaline phosphate. In order to accomplish these goals, 5 samples were used. Each sample contained different volumes of 0.2 m MPNPP (p-nitrophenylphosphate) and 0.2 M Tris-Hcl at a pH of 8.0. To each sample 0.2 mL of the enzyme studied (Alkaline Phosphatase) were added upon insertion on the spectrophotometer apparatus. With intervals of 20 seconds their absorbance at a wavelength of 410 nm was recorded at time frame of 2 minutes for each solution.…

John Cole Hairston Biology Lab 2107 W 7:00 October 14th, 2015 Analysis of Consumer Products with Unknown Protein Concentrations Using Spectrophotometry Abstract Consumer products typically give detailed information about their ingredients and nutrients. However, the concentration of a specific nutrient may not always be clear from just the label information alone, and in some cases the product labels may not be accurate. Thus, it may be hard to compare the concentration level of a certain nutrient, such as protein, amongst different products.…

Proteins are complex molecules that serve a variety of functions within the body. Proteins are made up of subunits called amino acids. There are several different types of amino acids such as lysine, aspartic acid, and histidine. Each type of amino acids have their own specific group called R-groups attached to them, this causes them to behave chemically different. The group on the amino acid histidine cause the amino acid to be basic; this feature of histidine makes it have a specific purpose in proteins, such as binding other molecules or atoms to a protein and also participating in a catalytic triad.…

Introduction Affinity Chromatography, also referred to as biospecific chromatography, is a technique of partitioning rooted in biological affinity (Dean). This practice capitalizes on complex interactions such as bond formation between enzymes and substrate, or more broadly: macromolecules and ligands. Purification is at the center of the use of affinity chromatography. In general, samples may be run through a gel that contains a corresponding component that binds with the sample at certain conditions. These conditions vary from acidity level, contending ligands, to ionic strength (Dean).…

Two chromatographic methods were developed for the assay of the FDA approved lozenges containing dextromethorphan hydrobromide (DXT) and menthol (MNT). The first was a green HPTLC method which depends on using Merck HPTLC aluminum sheets of silica gel 60 F254 and methanol-ammonia (10:0.1, v/v) as mobile phase. This was followed by densitometric measurements of the spots which were retained at 0.28 ± 0.01 for DXT and 0.76 ± 0.02 for MNT scanned at 210 nm. The method was found linear over the concentration ranges of 0.1 – 1 μg.band−1 for both DXT and MNT with values of correlation coefficients exceeding 0.999. The other method was RP-HPLC method with stability indicating merits at which Agilent 5 HC-C18 (2) 150 x 4.6mm was used as stationary…

Specific activity is useful to know for a protein as it is a good indicator of an enzyme 's purity and quality. The Bradford assay measures total protein in the assay, and was calculated as 4.2mg/mL. The result was calculated from a set of four concentrations, of which there was a standard deviation of 1.3mg/mL and a standard error of 0.66mg/mL. These are relatively large values, perhaps indicating that there was error during the dilution of 1S. As the big increase in undiluted concentration is between 1S2 and 1S4, this indicates error in the dilution of 1S4 from 1S2.…
